摘要
目的探讨适当改进经典四氯化碳模型复制方法。方法采用适当改进的40%(体积比)四氯化碳腹腔注射造模,造模结束后检测谷丙转氨酶、谷草转氨酶活性、HPC3、C4、LN、HA含量;通过Mallory染色观察大鼠肝组织病理学变化。结果模型组肝湿重、肝指数、HPC3、C4、LN、HA含量以及肝纤维化程度均较正常组明显升高(P<0.01)。结论本法能够成功建立大鼠肝纤维化模型,死亡率极低,从而提高了模型质量以及实验效率。
Objective To evaluate the improved rat liver fibrosis model by intraperitoneal injection with CCL4.Methods The mod-el group of rats was established by intraperitoneal injection with 40% CCL4.(volume ratio).Then the activity of serum alanine aminotrans-ferase,aspartate aminotransferase and the liver fibrosis indexes in serum of HPC3,C4,LN,HA content were detected,and the pathological changes of liver were observed with Mallory staining.Results The wet liver weight,liver index,HPC3,C4,LN,HA content and the degree of liver fibrosis of model group were increased significantly than those of the normal group(P<0.01).Conclusion The model of liver fibrosis is established successfully with this method,and the mortality rate is reduced,thus the experiment model quality and efficiency can be im-proved.
