摘要
目的:探讨运腹通经推拿法改善胰岛素抵抗(IR)肥胖大鼠模型的IR效果及相关作用影响,初步确定运腹通经推拿法通过对IR肥胖模型大鼠血清游离脂肪酸(FFA)、脂联素(ADPN)以及骨骼肌中沉默信息调节因子1(SIRT1)的调节,从而改善IR状态。方法:高脂饮食喂养8周,建立IR肥胖大鼠模型。将大鼠随机分为空白组、模型组、药物组和推拿组。高脂饲料喂养配合干预方法操作4周后,提取大鼠全血及股四头肌,对大鼠血清中FFA、ADPN含量,骨骼肌中SIRT1 mRNA表达水平进行测定。结果:推拿组大鼠血清中FFA含量显著低于模型组(P<0.05),ADPN含量则显著高于模型组(P<0.05),推拿组大鼠骨骼肌中SIRT1 mRNA表达水平显著高于模型组(P<0.05)。结论:运腹通经推拿法能够显著改善高脂饮食诱导IR大鼠模型,其作用机制可能与该手法调节血清中FFA、ADPN水平及骨骼肌中SIRT1表达水平有关。
Objective: To explore the related effect of the promoting channel abdominal massage on improving the insulin resistance(IR) of IR rats. To preliminary determined whether this therapy has a regulatory effect on serum free fatty acid(FFA), adiponectin(ADPN) and SIRT1 in skeletal muscle in IR rat model, so as to improve IR status. Methods: IR rat model was established after 8 weeks of high fat diet. The rats were randomly divided into control group, IR model group(model group), IR model+drug intervention group(drug group) and IR model+massage intervention group(massage group). After 4 weeks of operation with high-fat feed and intervention method, the whole blood and quadriceps femoris were extracted from the rats to measure the FFA, ADPN level in the serum and SIRT1 mRNA expression level in skeletal muscle. Results: The level of FFA in the serum of the massage group was significantly lower than that of the model group(P<0.05), the content of ADPN in serum was significantly higher than that in the model group(P<0.05), the mRNA expression of SIRT1 in skeletal muscle of the massage group was slightly higher than that in the model group(P<0.05). Conclusion: The promoting channel abdominal massage treatment can significantly improve the status in high-fat diet-induced IR rat model, and its mechanism may be related to the regulation of serum FFA and ADPN levels by this manipulation, so as to affect the expression level of SIRT1 in skeletal muscle.
