摘要
目的研究miRNA-380-5p对乳腺癌细胞增殖和凋亡的影响.方法收集2018年1-10月该院手术切除并经病理诊断确诊为乳腺癌的病理新鲜标本70例为研究组,同时选取病灶5 cm以上的癌旁正常组织为对照组.检测细胞系中miR-380-5p的表达量,并检测miR-380-5p在乳腺癌患者体内核因子蛋白90 (NF90)的表达情况,采用细胞计数法和平板克隆实验分别检测过表达miR-380-5p后乳腺癌细胞活力和增殖能力,采用流式细胞仪检测细胞凋亡情况.结果肿瘤组织和癌旁组织miR-380-5p表达量比较,差异有统计学意义(P<0.05).乳腺癌BT-549,MDA-MB-231,MCF-7,正常上皮细胞MCF-10A中miR-380-5p表达量比较,差异有统计学意义(P<0.05).转染后研究组NF90 mRNA表达量与阴性对照组比较,差异有统计学意义(P<0.05).进行qPCR检测后,在第4天、第5天MCF-7细胞的增殖能力显著高于阴性对照组,差异有统计学意义(P<0.05).结论miRNA-380-5p对乳腺癌细胞增殖具有抑制作用,对乳腺癌细胞凋亡具有促进作用.
Objective To research the effect of miRNA-380-5 p on proliferation and apoptosis of breast cancer cells.Methods Seventy samples of breast cancer tissue after surgery and pathological examination were collected from the hospital from January to October in2018 as study group,the normal breast tissue samples with distance to breast cancer>5 cm were collected as control group.The expression levels of miR-380-5 p and NF90 were detected,cell counting method and panel cloning experiment were used to detect cell activity and proliferation ability of breast cancer cells with overexpression of miR-380-5 p,flow cytometry was used to detect cell apoptosis.Results There was statistically significant difference in expression level of miR-380-5 p between study group and control group(P<0.05).There were statistically significant differences in expression level of miR-380-5 p between breast cancer BT-549,MDA-MB-231,MCF-7 cells and normal breast MCF-10 A cells(P<0.05).After transfection,there was statistically significant difference in expression level of NF90 mRNA between study group and control group(P<0.05).q PCR showed that the proliferation ability of MCF-7 cells on the fourth day and the fifth day in study group was statistically significantly higher than that in control group(P<0.05).Conclusion miRNA-380-5 p can inhibit proliferation and promote apoptosis of breast cancer cells.
