摘要
目的探讨尼克酰胺核糖(NR)对2型糖尿病(T2DM)小鼠模型的血糖调节能力的影响。方法29只雄性C57BL/6J小鼠分为对照组(n=8)、模型组(n=6)、低剂量NR组[400 mg/(kg·d)](n=8)、高剂量NR组[800 mg/(kg·d)](n=7)。对照组全程饲喂对照饲料,后三组利用高脂喂养和多次低剂量腹腔注射链脲佐菌素共同诱导2型糖尿病模型,在造模成功后,NR/生理盐水每日通过灌胃的方式给予。采用糖耐量实验(IPGTT)、胰岛素抵抗实验(IPITT)、空腹血糖和空腹胰岛素水平反映小鼠的血糖调节能力。结果NR组小鼠体重低于模型组,但差异无统计学意义(P>0.05);模型组小鼠与低剂量NR组和高剂量NR组小鼠的空腹血糖差异无统计学意义(P>0.05)。糖耐量实验的数据显示,低剂量NR组和高剂量NR组小鼠糖耐量实验血糖曲线下面积(AUC)均低于模型组,差异有统计学意义(P<0.05)。在胰岛素抵抗实验中,低剂量NR组和高剂量NR组与模型组的血糖AUC比较,差异无统计学意义(P>0.05)。空腹胰岛素检测结果显示,低剂量NR组和高剂量NR组的空腹胰岛素水平与模型组比较有升高趋势,但差异无统计学意义(P>0.05)。结论NR对T2DM小鼠的糖耐量受损有一定改善作用,可能是通过修复胰岛β细胞的分泌功能,在某种程度上提高空腹胰岛素的水平发挥其保护效应所致。
Objective To explore the effect of nicotinamide riboside(NR)on reversing glucose metabolic disorder of type 2 diabetes mellitus(T2 DM)mice.Methods 29 C57 BL/6 J mice were divided into four groups:control group(n=8),model group(n=6),low-dose NR group[400 mg/(kg·d)](n=8),and high-dose NR group[800 mg/(kg·d)](n=7).The control group was fed chow diet until the end of the experiment,whereas the other 3 groups were fed a high fat diet along with 4 times low-dose of streptozotocin(STZ)injection.After successfully inducing T2 DM in groups other than control,nicotinamide riboside solution or PBS were given by gavage every day.The intraperitoneal injection glucose tolerance test(IPGTT),intraperitoneal injection insulin tolerance test(IPITT),fasting blood glucose and fasting insulin level were used to evaluate the glucose regulation ability of mice.Results Intervention of NR(both low-dose and high-dose)did not result in significant change in body weight compared to control and model group(P>0.05).According to the IPGTT,area under the curve(AUC)of low-dose NR group and high-dose NR group were significantly lower than that of the model group(P<0.05).However,in the IPITT,there was no significant difference in AUC among model group,low-dose NR group and high-dose NR group(P>0.05).The fasting insulin showed an increasing trend in low-dose NR group and high dose NR group,but with no statistical significance(P>0.05).Conclusion NR could improve the impaired glucose tolerance of T2 DM mice,which might through improve the fasting insulin level.
