摘要
目的:探讨OTUD5在宫颈癌组织的表达及OTUD5过表达对宫颈癌Hela细胞系增殖的影响。方法:采用免疫组化及RNA聚合酶链反应(reverse transcription-polymerase chain reaction,RT PCR)技术检测宫颈癌组织及癌旁组织的OTUD5表达;Western blotting检测人宫颈癌Hela细胞中OTUD5蛋白的表达情况;构建过表达OTUD5的Hela细胞系,采用CCK-8法和平板克隆技术检测过表达组及对照组细胞增殖能力的变化。结果:OTUD5阳性表达主要着色于细胞浆,在宫颈癌组织中,高表达率为38.3%(23/60),在癌旁组织中高表达率为21.7%(13/60),两组之间行χ^2检验,差异有统计学意义(χ^2=3.968,P=0.046);在新鲜癌组织及癌旁组织之间,OTUD5 mRNA表达水平明显高于癌旁组织,差异均有统计学意义(P<0.05)。pcDNA3.1转染后细胞内OTUD5蛋白相对表达水平为0.435±0.048,pcDNA3.1-OTUD5转染后细胞内OTUD5蛋白相对表达水平为0.224±0.139,两组间差异有统计学意义(t=-3.162,P=0.012)。CCK-8法及平板克隆实验显示,过表达OTUD5组较对照组增殖及克隆能力均增强(P<0.05)。结论:OTUD5在宫颈癌中高表达,并能促进Hela细胞的克隆及增殖。
Objective To investigate the effect of OTUD5 on cervical cancer tissue and OTUD5 overexpression on the proliferation of cervical cancer Hela cell line.Method Immunohistochemistry and reverse transcription-polymerase chain reaction(RT-PCR)were used to detect the expression of OTUD5 in cervical cancer tissues and adjacent tissues.The expression of OTUD5 protein in human cervical cancer Hela cells was detected by Western blotting.The Hela cell line overexpressing OTUD5 was constructed,and the cell proliferation ability and plate clone change of the overexpression group and the control group were detected by CCK-8 method.Results OTUD5 positive expression was mainly stained in cytoplasm.In cervical cancer tissues,the high expression rate was 38.3%(23/60),and the high expression rate in adjacent tissues was 21.7%(13/60).The square test showed statistical difference(χ^2=3.968,P=0.046).The expression level of OTUD5 mRNA was significantly higher in fresh cancer tissues and adjacent tissues than in adjacent tissues(P<0.05).The relative expression level of OTUD5 protein in transfected cells was 0.435±0.048 after transfection of pcDNA3.1,and the relative expression level of OTUD5 protein in transfected cells was 0.224±0.139 after transfection.The difference between the two groups was statistically significant(t=-3.162,P=0.012).CCK-8 assay and plate cloning experiments showed that the overexpressed OTUD5 group had enhanced proliferation and clonal ability compared with the control group(P<0.05).Conclusion OTUD5 is highly expressed in cervical cancer and can promote the cloning and proliferation of Hela cells.
