摘要
目的研究尼妥珠单抗(h-R3)和西托昔单抗(C225)对食管癌患者放射增敏作用的影响。方法体外培养人食管癌细胞株TE-13,分为对照组(单纯X线照射干预)、h-R3组(X线照射+h-R3干预)及C225组(X线照射+C225干预)。完成后收集细胞,采用免疫印迹法(Western blot)检测EGFR信号通路相关蛋白水平,应用克隆形成实验分析h-R3与C225对食管癌细胞株放疗敏感性的影响。分别采用单溶液细胞增殖分析(MTS)法和流式细胞术检测各组细胞增殖率和凋亡率。结果Western blot显示C225组p-EGFR、p-MAPK表达均低于对照组和h-R3组(P<0.05)。克隆形成实验显示h-R3与C225作用于食管癌细胞的放射增敏比(SER)分别为1.35和1.50,C225组平均致死量(D0)、准阈剂量(Dq)、外推值(N)及2 Gy照射后存活分数(SF2)均低于对照组和h-R3组(P<0.05)。C225组细胞增殖率显著低于对照组和h-R3组,细胞凋亡率显著高于对照组和h-R3组,差异均有统计学意义(P<0.05)。三组食管癌TE-13细胞株G2/M与S期细胞周期分布比率差异显著(P<0.05),其中C225组G2/M期细胞比例显著高于对照组和h-R3组,S期细胞比例显著低于对照组和h-R3组(P<0.05)。结论h-R3与C225对食管癌放疗均具有增敏效果,且C225增敏作用较h-R3作用更为显著,这可能与其抑制EGFR及下游蛋白表达有关。
Objective To study the vitro experiment of radiosensitizing effect of nitorzumab(h-R3)and cetuximab(C225)on esophageal cancer.Methods The human esophageal cancer cell line TE-13 were cultured in vitro,they were divided into control group(X-ray intervention),h-R3 group(X-ray+h-R3 intervention)and C225 group(X-ray+C225 intervention).The cells were collected after completion,and the EGFR signal pathway related proteins were detected by Western blot,the effect of h-R3 and C225 on radiosensitivity of esophageal cancer cell line were analyzed by clonogenesis.The cell proliferation rate and apoptosis rate were detected by single solution cell proliferation analysis(MTS)and flow cytometry.Results The Western blot showed that the expression of p-EGFR and p-MAPK in C225 group were lower than those in control group and h-R3 group.The sensitizatio enhancement ratio(SER)of h-R3 and C225 on esophageal cancer cells were 1.35 and 1.50,respectively,the mean lethal dose(D0),quasithreshold dose(Dq),extrapolation value(N),surviving fraction at 2 Gy(SF2)in C225 group were lower than those of the contral group and h-R3 group.The cell proliferation rate of C225 group was significantly lower than that of the control group and h-R3 group,and the apoptosis rate was significantly higher than that of the control group and h-R3 group,the difference were statistically significant(P<0.05).There were significant difference between the G2/M and S-phase cell cycle distribution ratio of TE-13 cell lines in three groups(P<0.05),the G2/M-phase cell ratio of C225 group was significantly higher than those of the control group and h-R3 group,and the S-phase cell ratio was significantly lower than those of the control group and h-R3 group(P<0.05).Conclusion The h-R3 and C225 have sensitization effect on esophageal cancer radiotherapy,and C225 has more significant sensitization effect than h-R3,which may be related to the inhibition of EGFR and downstream protein expression.
