摘要
目的分析在鼻咽癌患者中血清microRNAlet-7a与其治疗、预后的相关性。方法从收治的鼻咽癌患者中,随机选择73例患者作为研究组;另随机选择73例同期行体检的健康人作为对照组。收集两组患者的血清,应用ELISA法检测两组患者血清中的碱性成纤维生长因子、血管内皮生长因子的表达水平。收集研究组患者的癌组织与对照组的正常鼻咽组织,应用RT-PCR法检测两组患者组织中的let-7a、Lin28的microRNA表达水平以及C-myc、HMGA2、H-Ras的mRNA表达水平,应用western-blot法检测两组患者组织中的HMGA2、血管内皮生长因子、p-PI3K、pAkt表达水平。患者结束治疗5年内,对患者进行随访,统计患者的5年生存率。结果对照组组织中的let-7a microRNA表达水平显著高于研究组(P<0.05),而研究组组织中lin28 mRNA(lin28a、lin28b)水平显著高于对照组(P<0.05);研究组组织中的碱性成纤维生长因子、血管内皮生长因子表达水平显著低于对照组(P<0.05);研究组组织中HMGA2的mRNA与蛋白表达水平均显著高于对照组(P<0.05),而对照组的5年生存率显著高于研究组患者(P<0.05);研究组患者组织中的血管内皮生长因子、p-PI3K、pAkt蛋白表达水平显著高于对照组(P<0.05)。结论应用血清中的microRNAlet-7a水平与其他生化分子预测鼻咽癌患者的病情及预后生活质量具有比较好的效果,为患者赢得尽可能多的治疗时间,能够有效提高患者的5年生存率。
Objective To analyze the correlation between serum microRNAlet-7a and its treatment and prognosis in patients with nasopharyngeal carcinoma.Methods 73 patients with nasopharyngeal carcinoma were randomly selected as the study group;another 73 healthy patients who were examined during the same period were randomly selected as the control group.The serum of the 2 groups of patients was collected,and the expression levels of basic fibroblast growth factor and vascular endothelial growth factor in the serum of the 2 groups were detected by ELISA.The cancer tissues of the study group and normal nasopharyngeal tissues of the control group were collected,and the expression levels of microRNAs of let-7a and Lin28 and the mRNA expressions of C-myc,HMGA2,and H-Ras in the tissues of the 2 groups were detected by RT-PCR.The levels of HMGA2,vascular endothelial growth factor,p-PI3K and pAkt in the tissues of the 2 groups of patients were detected by western-blot method.Within 5 years after the end of treatment,the patients were followed up to calculate the 5-year survival rate of the patients.Results The expression level of let-7a microRNA in the control group was significantly higher than that of the study group(P<0.05),while the level of lin28 mRNA(lin28a,lin28b)in the study group was significantly higher than that of the control group(P<0.05);The expression levels of basic fibroblast growth factor and vascular endothelial growth factor were significantly lower than those of the control group(P<0.05);the mRNA and protein expression levels of HMGA2 in the study group were significantly higher than those of the control group(P<0.05),and 5-year survival rate in the control group was significantly higher than that of the study group(P<0.05);the levels of vascular endothelial growth factor,p-PI3K,and pAkt proteins in the tissues of the study group were significantly higher than those of the control group(P<0.05).Conclusion The use of serum microRNAlet-7a levels and other biochemical molecules to predict the condition and quality
