摘要
目的探讨中药缓压乐复方制剂对应激性高血压(stress induced hypertension,SIH)大鼠心肌组织受磷蛋白(Phospholamban,PLB)磷酸化水平的调节作用。方法用复合应激因素建立SIH大鼠模型的基础上,以中药缓压乐复方制剂进行干预。采用实时荧光定量PCR(RT-qPCR)和免疫组织化学方法分别检测SIH模型组、正常组、药物干预组和阴性对照组大鼠心肌组织内源性抑制剂1(endogenous inhibitor 1,I-1)基因mRNA表达以及PLB蛋白的磷酸化水平。结果与正常组相比,模型组大鼠心肌组织I-1基因mRNA表达水平明显下调(P<0.05),用药物干预后明显上调(P<0.05);与正常组相比,模型组大鼠心肌组织PLB蛋白磷酸化水平明显下调(P<0.05),而用药物干预后明显上调(P<0.05)。结论中药缓压乐复方制剂可能通过PLB磷酸化水平的上调,增强SERCA2a蛋白的活性,从而增加肌质网对Ca^2+的重吸收。
Objective To investigate the regulating effect of traditional Chinese medicine Huan Ya Le on the phosphorylation level of Phospholamban(PLB)in cardiac muscle tissue of stress induced hypertension(SIH)rats.Methods The rat model of SIH was established by combined stress,and on this basis,traditional Chinese medicine Huan Ya Le was given for intervention.mRNA expression of endogenous inhibitor-1(I-1)gene and phosphorylation level of PLB in the SIH model group,normal group,drug intervention group and negative control group were determined by real-time fluorescence quantitative PCR(RT-qPCR)and immunohistochemistry,respectively.Results Compared with the normal group,the mRNA expression level of I-1 gene in the model group was significantly down-regulated(P<0.05)and significantly up-regulated after the drug intervention(P<0.05).Compared with the normal group,the phosphorylation level of PLB protein in the model group was significantly down-regulated(P<0.05),and significantly up-regulated after the drug intervention(P<0.05).Conclusion Traditional Chinese medicine Huan Ya Le may increase the level of phosphorylation of PLB to enhance the activity of SERCA2a protein to normal,thereby increasing the Ca^2+reabsorbtion of sarcoplasmic reticulum.
