利用高速逆流色谱技术制备灵芝酮三醇 被引量:4

Preparative Isolation of Ganodermanontriol from Ganoderma lucidum by High-speed Countercurrent Chromatography

摘要 以沪农灵芝(Ganoderma lucidum)1号子实体为原料,经过87%乙醇提取、D101大孔树脂富集、乙醇梯度洗脱获得弱极性三萜组分。采用高速逆流色谱技术,以正己烷∶乙酸乙酯∶甲醇∶水(V∶V∶V∶V=5∶6∶5∶5)为溶剂体系,对该三萜组分中灵芝酮三醇的制备工艺进行研究,在转速900 r·min^-1、流速3 mL·min^-1条件下,一次上样400 mg灵芝弱极性三萜组分,可得到纯度为91.6%的灵芝酮三醇26.8 mg。 Ganodermanontriol was purified from fruiting bodies of Ganoderma lucidum(Hunong No.1) following a series of steps, including extraction by 87% ethanol, condensation by macroporous adsorption resin D101, gradient elution by ethanol to obtain the weak polar triterpenes fraction(WTF), and finally preparation of the target compound from WTF by high-speed countercurrent chromatography(HSCCC). For HSCCC, the solvent system was determined to be a mixture of N-hexane,ethyl acetate, methanol and water(V∶V∶V∶V=5∶6∶5∶5);the separation condition was set as rotation at 900 rpm and flow rate of 3.0 mL·min^-1. Using the above mentioned HSCCC condition, a single loading amount of 400 mg WTF yielded 26.8 mg ganodermanontriol with a purity of 91.6%.
