摘要
为探讨饲料中益生菌和不同水平壳聚糖对黄金鲫非特异性免疫力和抗病力的影响,选取480尾初始体重为(58.94±4.29) g的健康黄金鲫随机分为6组,每组4个重复,每个重复20尾鱼,分别投喂基础日粮(C1组),益生菌(8 g/kg)饲料(C2组),益生菌(8 g/kg)+0.25%壳聚糖饲料(T1组),益生菌(8 g/kg)+0.50%壳聚糖饲料(T2组),益生菌(8 g/kg)+0.75%壳聚糖饲料(T3组),益生菌(8 g/kg)+1.00%壳聚糖饲料(T4组),试验周期为30 d。结果表明:(1)与C1组相比,T2和T3组血清、肝胰脏、脾脏、头肾、中肾、脑和鳃中超氧化物歧化酶(SOD)活性分别提高7.07%~40.47%和5.75%~46.05%,效果显著(P <0.05);(2)T2组肝胰脏、脾脏、脑和鳃中过氧化氢酶(CAT)活性分别增长14.62%、42.55%、26.89%和27.68%,效果显著(P <0.05),T3组血清、头肾、中肾中CAT活性最大(P <0.05),分别为23.98、5.43和2.46;(3)T4组血清中谷胱甘肽(GSH)含量最高,为24.95,其余组织中GSH含量最大值出现在T3组(P <0.05);(4)T2组脾脏和脑中谷胱甘肽过氧化物酶(GSH-Px)活性显著升高,分别为对照组的1.30和1.32倍(P <0.05),其余组织酶活性最大值出现在T3组(P <0.05);(5)T2~T4组各组织中丙二醛(MDA)含量具有不同程度的降低,除鳃之外,均在T4组达到最低值(P <0.05);(6)T3组血清、肝胰脏、脾脏和脑中NO含量达到最大值(P <0.05),T4组头肾、中肾和鳃中NO含量达到最大值(P <0.05);(7)T3、T4组肝胰脏中谷草转氨酶(GOT)和谷丙转氨酶(GPT)活性较对照组降低71%~89%(P <0.05);(8)T2、T3组累计死亡率分别降低40%和60%,C2组与C1组变化不明显(P> 0.05)。上述结果表明,饲料中添加壳聚糖可有效提高黄金鲫非特异性免疫力和抗病力,且适宜添加水平为0.5%~0.75%。
The present study aimed to evaluate the effects of probiotics and chitosan on non-special immunity and disease resistance of the golden Carassius auratus.Four hundred and eighty golden Carassius auratus juveniles,with the initial body weight of(58.94±4.29)g,were randomly divided into six groups,with four replicates per group and 20 fish in each replicate.The six groups were fed with base diet(C1 group),diet with 8 g/kg probiotics(C2 group),diet with 8 g/kg probiotics and 0.25% chitosan(T1 group),diet with 8 g/kg probiotics and 0.50% chitosan(T2 group),diet with 8 g/kg probiotics and 0.75% chitosan(T3 group),diet with 8 g/kg probiotics and 1.0% chitosan(T4 group).The results showed that:(1) Compared with C1 group,the superoxide dismutase(SOD) activities in the serum,hepatopancreas,spleen,head kidney,mid kidney,brain and gill of T2 and T3 groups was significantly increased by 7.07% ~ 40.47% and 5.75% ~ 46.05%,respectively(P < 0.05);(2) The catalase(CAT) activities in the hepatopancreas,spleen,brain and gill of T2 group was significantly improved by 714.62%,42.55%,26.89% and 27.68%,respectively(P < 0.05).CAT activities in serum,head kidney and mid kidney of T3 group reached to maximum,which was 23.98,5.43 and 2.46(P < 0.05);(3) The highest glutathione(GSH) content in serum was obtained in T4 group,and maximum GSH content(about24.95) in other tissues was obtained in T3 group(P < 0.05);(4) The T2 group displayed significant increases in glutathione peroxidase(GSH-Px) activities in the spleen and brain,which was 1.30 and 1.32 times of that in the control group(P < 0.05),and maximum GSH-Px activities in other tissues was obtained in T3 group(P <0.05);(5) The malondialdehyde(MDA) content in tissues of T2 ~ T4 groups decreased in various degrees,reaching the lowest value in T4 group except in gills;(6) NO content in serum,hepatopancreas,spleen and brain reached to the maximum(P < 0.05);(7) Compared with the control group,the glutamic oxaloacetylase(GOT) and alanice aminotransferase(GPT) activities in the hepatopancreas o
