摘要
目的对比不同机型不同检测试剂检测人体CD4^+T淋巴细胞数值,为建立快速、稳定且敏感性高的检测平台提供依据。方法对比BD的3个型号机器(Calibur,Count和Presto)及艾森ACEA NovoCyte流式细胞仪平行检测111份艾滋病感染者/病人(HIV/AIDS)血样中CD4+T淋巴细胞计数结果,以统计学软件评估检测结果的一致性和差异性;利用单平台对比不同试剂对CD4^+T淋巴细胞计数检测结果的影响。结果3种BD机型的CD4^+T淋巴细胞计数结果差异无统计学意义(Pearson r=0.962,0.977和0.978,P>0.05),但Presto检测步骤少、操作简单;在BD Calibur检测平台上,BD试剂和同生时代试剂检测结果的Pearson r=0.904,差异有统计学意义(P<0.05);在艾森平台上,不同试剂检测结果差异无统计学意义(Pearson r=0.983,0.975和0.970,P>0.05)。结论BD各型号机器对HIV/AIDS的CD4^+T淋巴细胞检测结果有高度的一致性,但对于国产试剂的兼容性不如艾森流式细胞仪。
Objective To compare CD4^+T lymphocyte values of human body by combining different test reagents with different models,provide a basis for establishing a rapid,stable and sensitive test platform.Methods The results of CD4^+T lymphocyte count in the blood samples of 111 HIV infectors/AIDS patients(HIV/AIDS)were compared by three BD machines(Calibur,Count and Presto)and parallel detection(ACEA NovoCyte).The consistency and difference in the results were evaluated with statistical software.The effects of different reagents on CD4^+T lymphocyte count on a single platform were compared.Results There was no statistical difference in CD4^+T lymphocyte count results among the three BD models(Pearson correlation coefficients of 0.962,0.977 and 0.978,respectively,P>0.05).However,Presto detection was of few procedures and simple operation.The Pearson coefficient of BD reagent and syngenetic reagent on BD Calibur detection platform was 0.904,with statistical difference(P<0.05).On ACEA platform,there was no statistical difference in the detection results of different reagents(Pearson correlation coefficients of 0.983,0.975 and 0.970,P>0.05).Conclusions The detection results of CD4^+T lymphocyte of HIV/AIDS by BD machines are highly consistent,but the compatibility of domestic reagent is not as good as that of ACEA flow cytometry.
