摘要
为探讨从绶草中分离得到的新化合物spiranthesphenanthrene A对人胃癌AGS细胞凋亡的作用及可能机制,利用MTT法检测spiranthesphenanthrene A对AGS细胞活力的影响;流式细胞术检测细胞凋亡情况;Western blotting检测凋亡相关蛋白表达水平。结果表明:spiranthesphenanthrene A呈剂量和时间依赖性地抑制AGS细胞活力(P<0.05);与空白对照组相比, spiranthesphenanthrene A可显著诱导AGS细胞凋亡,且呈剂量依赖性(P<0.05)。进一步研究表明, spiranthesphenanthrene A可显著上调凋亡相关蛋白Cleaved-caspase-3、Cleaved-caspase-9、Bax和Cytochrome C (Cyt-C)的表达,并下调Bcl-2的表达(P<0.05)。这一研究提示spiranthesphenanthrene A能抑制AGS细胞活力,并通过内源性线粒体途径诱导细胞凋亡。
The purpose of the study was to investigate the effect of spiranthesphenanthrene A, a new compound isolated from Spiranthes sinensis, on the apoptosis of human gastric cancer AGS cells and its mechanism. The effect of spiranthesphenanthrene A on the viability of AGS cells was detected by a MTT assay. Cell apoptosis was measured by flow cytometry. The expression levels of apoptosis-related proteins were determined by Western blotting. The results showed that spiranthesphenanthrene A inhibited the viability of AGS cells in a dose-and time-dependent manner(P<0.05). Compared with the control group, spiranthesphenanthrene A significantly induced the apoptosis in AGS cells in a dose-dependent manner(P<0.05). In addition, spiranthesphenanthrene A could significantly upregulate the protein expression levels of Cleaved-caspase-3, Cleaved-caspase-9, Bax and Cytochrome C(Cyt-C) and downregulate those of Bcl-2(P<0.05). The present study indicates that spiranthesphenanthrene A inhibits cell viability and induces cell apoptosis via mitochondria-mediated intrinsic pathway in AGS cells.
