微小RNA-122在妊娠期糖尿病中的表达及意义 被引量:11

Expression of miR-122 in patients with gestational diabetes mellitus and its clinical significance

摘要 目的探讨微小RNA-122(miR-122)在妊娠期糖尿病(GDM)患者血清和胎盘组织中的表达情况及意义。方法选择2018年1月至2019年6月在南方医科大学附属东莞人民医院进行产检并住院分娩的GDM患者50例(GDM组)、健康孕产妇35例(对照组)。采用实时荧光定量PCR检测各组血清和胎盘组织中miR-122的表达水平。构建胰岛素抵抗细胞模型,利用脂质体将miR-122模拟物转染至细胞,应用Western blot检测AMPK蛋白的表达水平。结果GDM组患者血清和胎盘组织中miR-122的表达水平(分别为1.18±0.45、1.11±0.48)高于对照组(分别为0.89±0.36、0.81±0.39),差异有统计学意义(P<0.05)。miR-122的表达水平与胰岛素抵抗指数呈正相关(P<0.05)。体外细胞实验显示,miR-122在软脂酸诱导胰岛素抵抗HepG2细胞中表达上调,体外转染miR-122模拟物可下调AMPK蛋白表达水平,并降低葡萄糖利用率。结论miR-122在GDM患者中的表达增高,其可能通过靶向调控AMPK信号通路参与GDM的病理生理过程。 Objective To investigate the expression of miR-122 in patients with gestational diabetes mellitus(GDM)and its clinical significance.Methods A total of 50 pregnant women with GDM and 35 healthy controls were recruited from January 2018 to June 2019 at Dongguan People’s Hospital of Southern Medical University.The expression levels of miR-122 in serum and placenta tissue were determined by real-time quantitative PCR.The insulin resistance model was induced by plamitic acid.The miR-122 mimics were transiently transfected into insulin resistant cells,and the AMPK gene expression was analyzed by Western blot.Results Higher miR-122 expression was found in GDM(1.18±0.45,1.11±0.48)cases compared to controls(0.89±0.36,0.81±0.39)(P<0.05).The expression levels of miR-122 were positively correlated with the insulin resistance index(P<0.05).Up-regulation of miR-122 was found in insulin resistant HepG2 cells,and miR-122 mimics transfected in vitro inhibited AMPK expression and decreased glucose disposal.Conclusion The expression of miR-122 was increased in patients with GDM,which indicates that miR-122 might be involved in the pathophysiological process of GDM through targeted regulation of AMPK signal pathway.
