摘要
目的评价电针预处理对小鼠脑缺血再灌注时海马核转录因子NF-E2相关因子2(Nrf2)/血红素加氧酶-1(HO-1)信号通路的影响。方法选择健康雄性C57BL/6小鼠120只,10~12周龄,体重25~30 g,采用随机数字表法分为4组(n=30):对照组(C组)、脑缺血再灌注组(I/R组)、电针无关穴位预处理+脑缺血再灌注组(S+I/R组)和电针百会穴预处理+脑缺血再灌注组(E+I/R组)。采用双侧颈总动脉阻闭法制备小鼠脑缺血再灌注损伤模型。E+I/R组电针刺激百会穴30 min,选取疏密波,强度1 mA,频率2/15 Hz,1次/d,连续5 d,最后1次电针刺激结束后24 h时建立脑缺血再灌注损伤模型。S+I/R组针刺百会穴旁2 mm,其余处理同E+I/R组。于再灌注24和48 h时行神经行为学评分,然后处死小鼠,取海马组织,HE染色后观察海马CA1区病理学结果,分别采用Western blot法和RT-PCR法测定核蛋白Nrf2及其mRNA及总蛋白HO-1、醌氧化还原酶1(NQO-1)及其mRNA的表达水平。结果与C组相比,I/R组、S+I/R组和E+I/R组神经行为学评分升高,核蛋白Nrf2及其mRNA和总蛋白HO-1、NQO1及其mRNA表达上调(P<0.05);与I/R组相比,E+I/R组神经行为学评分降低,核蛋白Nrf2及其mRNA和总蛋白HO-1、NQO1及其mRNA表达上调(P<0.05),S+I/R组上述各指标差异无统计学意义(P>0.05)。与S+I/R组比较,E+I/R组神经行为学评分降低,核蛋白Nrf2及其mRNA和总蛋白HO-1、NQO1及其mRNA表达上调(P<0.05)。结论电针预处理减轻小鼠脑缺血再灌注损伤的机制可能与激活海马Nrf2/HO-1信号通路有关。
Objective To evaluate the effect of electroacupuncture(EA)preconditioning on hippocampal nuclear factor erythroid 2-related factor 2(Nrf2)/heme oxygenase-1(Nrf2/HO-1)signaling pathway during cerebral ischemia-reperfusion(I/R)in mice.Methods A total of 120 healthy male C57BL/6 mice,aged 10-12 weeks,weighing 25-30 g,were divided into 4 groups(n=30 each)using a random number table method:control group(group C),cerebral I/R group(group I/R),preconditioning with EA at non-acupoint+cerebral I/R group(group S+I/R)and preconditioning with EA at Baihui acupoint+cerebral I/R group(group E+I/R).The cerebral I/R injury model was established by occlusion of bilateral common carotid arteries followed by reperfusion for 72 h in mice anesthetized with halothane or chloral hydrate.Group E+I/R received EA at Baihui acupoints with a sparse-dense wave at an intensity of 1 mA and a frequency of 2/15 Hz for 30 min once a day for 5 consecutive days,and then the cerebral I/R injury model was established at 24 h after the end of the last stimulation.Group S+I/R received EA at the points 2 mm lateral to the acupoints of Baihui,and the other treatments were similar to those previously described in group E+I/R.Neurobehavioral score was assessed at 24 and 48 h of reperfusion.Then 5 mice in each group were sacrificed,and the hippocampal tissues were obtained and stained with haematoxylin and eosin for examination of the pathological changes in hippocampal CA1 region and for determination of the expression of Nrf2 protein and mRNA in nucleoprotein and HO-1 and quinone oxidoreductase 1(NQO1)in total protein(by Western blot)and the expression of Nrf2 mRNA,HO-1 mRNA and NQO1 protein and mRNA in hippocampal tissues(using real-time polymerase chain reaction).Results Compared with group C,neurobehavioral score was significantly increased,and the expression of hippocampal Nrf2 protein and mRNA in nucleoprotein and HO-1 and NQO1 protein and mRNA in total protein was up-regulated in I/R,S+I/R and E+I/R groups(P<0.05).Compared with group I/R,neurobehavi
