摘要
目的观察升陷乌梅汤对哮喘大鼠激素干预模型气道炎症、气道重塑和肺功能的影响。方法50只SD雄性大鼠随机分为正常组、哮喘组、激素干预哮喘组、布地奈德组及升陷乌梅汤组,每组10只。采用卵蛋白致敏、雾化激发建立哮喘大鼠模型,并予正常组和哮喘组以外各组大鼠腹腔注射糖皮质激素干预,在此基础上予布地奈德组布地奈德混悬液[2 mL/(20 min·d)]雾化吸入,予升陷乌梅汤组升陷乌梅汤中药[0.81 g/(kg·d)]灌胃,均干预7周。运用动物肺功能分析系统检测分析0.3秒用力呼气容积(FEV0.3)、用力肺活量(FVC)、0.3秒内平均流速(FEV 0.3/FVC%)、呼气峰流速(PEF)、每分钟通气量(MVV)的变化;HE染色观察肺组织病理改变,并使用Image-Pro Plus 6.0软件测定气道平滑肌细胞核计数(NCASMc)、气道壁面积(Wat)及其百分比(%TAt),ELISA法检测肺泡灌洗液中转化生长因子-β(TGF-β)浓度;Western Blot法检测Ⅰ型胶原(CollagenⅠ)和Ⅲ型胶原(CollagenⅢ)蛋白表达水平。结果与正常组比较,哮喘组FEV0.3、FVC、FEV0.3/FVC%及PEF显著下降(P<0.05),NCASMc、Wat及%TAt显著升高(P<0.05),TGF-β、CollagenⅠ及CollagenⅢ蛋白表达显著升高(P<0.05)。与哮喘组比较,激素干预各组肺功能FEV0.3和FEV0.3/FVC%均升高(P<0.05),NCASMc和%TAt均降低(P<0.05),TGF-β、CollagenⅠ及CollagenⅢ蛋白表达均降低(P<0.05)。与激素干预哮喘组比较,升陷乌梅汤组部分肺功能FEV 0.3和FEV 0.3/FVC%升高(P<0.05),NCASMc及%TAt均有降低(P<0.05),CollagenⅠ及CollagenⅢ蛋白表达均降低(P<0.05)。结论糖皮质激素干预不能完全抑制哮喘大鼠气道炎症及气道重塑的发生,升陷乌梅汤可在激素干预基础上下调TGF-β等指标水平,进一步抑制气道炎症和气道重塑的发展,改善大鼠肺功能。
Objective To observe the effect of Shengxian Wumei Decoction(SXWMD)on airway in-flammation,airway remodeling,and lung function in asthmatic model rats treated with glucocorticoids.Methods Totally 50 SD male rats were randomly divided into the normal group,the asthma group,the glucocorticoids in-tervened asthma group,Budesonide group,and the SXWMD group,10 in each group.The asthma model was established by sensitization and stimulation with ovalbumin(OVA).Except for rats in normal and asthma groups,rats in the other groups were intraperitoneally injected with glucocorticoids.Based on the glucocorticoids intervention,rats in Budesonide group were administered with Budesonide by atomization[2 mL/(20 min·d)],and rats in SXWMD group were treated with intragastric administration of SXWMD[0.81g/(kg·d)].Lung function indices,including forced expiratory volume in 0.3 second(FEV0.3),forced vital capacity(FVC),FEV0.3/FVC%,peak expiratory flow(PEF),and minute ventilation volume(MVV)were detected using an AniRes2005 lung function test system.Histopathological changes of lung were observed by HE stai-ning.Nucleus count of airway smooth muscle cells(NCASMc),wall area of bronchial tube(Wat),and per-centage of tube wall area to total area(%TAt)were assessed with Image-Pro Plus 6.0.The expression level of transforming growth factor-β(TGF-β)was detected by ELISA.Western Blot was performed to determine the ex-pressions of typeⅠcollagen(CollagenⅠ)and typeⅢcollagen(CollagenⅢ).Results Compared with the normal group,lung function indices,incuding FEV0.3,FVC,FEV0.3/FVC%,and PEF,significantly de-creased(P<0.05),NCASMc,Wat,and%TAt significantly increased(P<0.05),and the levels of TGF-β,collagenⅠand collagenⅢsignificantly increased in the asthma group(P<0.05).Compared with the asth-ma group,FEV0.3 and FEV0.3/FVC%significantly increased(P<0.05),NCASMc and%TAt significantly decreased(P<0.05),and the expressions of TGF-β,collagenⅠand collagenⅢin lung tissues significantly decreased in all glucocorticoids intervened groups(
