摘要
目的探讨重建中气抗癌汤含药血清对人胃癌SGC7901细胞凋亡的影响及机制。方法Wistar大鼠随机分为空白组,重建中气抗癌汤低、中、高剂量(1.55、3.1、6.2 g/kg)组及5-氟尿嘧啶(5-FU)组制备含药血清。含药血清作用SGC7901细胞后,采用流式细胞术观察细胞凋亡率,梯度聚合酶链式反应(Polymerase Chain Reaction,PCR)检测B淋巴细胞瘤-2基因(B-cell lymphoma-2,Bcl-2)、BCL2-Associated X的蛋白质(Bax)、半胱氨酸天冬氨酸蛋白酶3(Caspase-3)mRNA活性;蛋白质印迹法(Western Blot)检测Bcl-2、Bax、Caspase-3蛋白表达情况。结果与对照组比较,经加味重建中气抗癌汤各浓度组处理后,各实验组SGC7901细胞的凋亡率显著高于对照组;重建中气抗癌汤处理后的促凋亡基因Caspase-3、Bax mRNA及蛋白表达均升高,抗凋亡基因Bcl-2的mRNA及蛋白表达均降低(P<0.05)。结论重建中气抗癌汤含药血清能够诱导胃癌SGC7901细胞凋亡,其机制可能与增强Caspase-3活性、上调Bax表达和下调Bcl-2表达有关。
Objective To investigate the effect and mechanism of the serum containing Reconstruction Zhongqi Anticancer Decoction(SCRZAD)on apoptosis of human gastric cancer SGC7901.Methods Wistar rats were randomly divided into blank group,low,medium and high doses(1.55,3.1,6.2 g/kg)group and 5-FU group to prepare drug-containing serum.The apoptotic rate of SGC7901 cells was observed by flow cytometry,the genes activity of Bcl-2,Bax and Caspase-3 mRNA was detected by gradient PCR,and the protein expression of Bcl-2,Bax and Caspase-3 was detected by Western Blot.Results Compared with the control group,the apoptotic rate of SGC7901 cells in each experimental group was significantly higher after treatment with SCRZAD at different concentrations than that in the control group,statistical differences were also found among different concentrations of drugs;Caspase-3,Bax genes and protein expression of apoptotic were increased after treatment with SCRZAD(P<0.05),while the expression of Bcl-2 gene and protein were decreased(P<0.05),statistical differences were also found among different concentrations of drugs.Conclusion The SCRZAD can induce apoptosis of SGC7901 cells,the mechanism may be related to the enhancement of Caspase-3 activity,up-regulation of Bax expression and down-regulation of Bcl-2 expression.
