摘要
目的研究连芩解毒方和万古霉素对耐甲氧西林金黄色葡萄球菌(MRSA)的体外联合抗菌作用。方法采用微量肉汤稀释法分别测定连芩解毒方和万古霉素对MRSA的最小抑菌浓度(MIC)值,利用棋盘法、时间-杀菌曲线评估连芩解毒方与万古霉素的抑菌作用。结果时间-杀菌曲线发现,连芩解毒方与万古霉素联合用药的抑菌活性提高了9.57(ΔLC24>2 log10 CFU·mL^-1),表明连芩解毒方与万古霉素对MRSA的抑制有协同作用。结论连芩解毒方对MRSA具有显著的抑制作用以及其与万古霉素联合产生了协同抗菌作用,为进一步研究连芩解毒方在体内的抑菌作用机制及更深层的挖掘其临床用途提供了理论基础。
Objective To determine the combined antibacterial effect of Lianqin Jiedu decoction and vancomycin on Methicillin-resistant Staphylococcus aureus(MRSA)in vitro.Methods Broth microdilution method was used to determine the minimum inhibitory concentration(MIC)of Lianqin Jiedu decoction and vancomycin on MRSA,and checkerboard method and time-kill curves were used to evaluate the antibacterial effect of Lianqin Jiedu decoction and vancomycin.Results The time-kill curves showed that the combined antibacterial activity of Lianqin Jiedu decoction and vancomycin increased by 9.57(ΔLC24>2 log10 CFU·mL^-1),indicating that Lianqin Jiedu decoction and vancomycin had a synergistic effect on MRSA.Conclusion Lianqin Jiedu decoction has a significant inhibitory effect on MRSA and a synergistic antibacterial effect with vancomycin,which provides a theoretical basis for further elucidating its antibacterial mechanism in vivo and further its clinical use.
