基因重组乳酸菌r-L.Lactis-VP2-RCK对IBDV母源抗体不同水平雏鸡的免疫效果

摘要 母源抗体是早期干扰活疫苗免疫效果的重要因素,通过研究商品蛋鸡法氏囊母源抗体的消长规律,并且在母源抗体达到最高峰的时候,观察免疫重组乳酸菌r-L.Lactis-VP2-RCK的免疫效果是否受到母源抗体的干扰。结果显示,母源抗体在3日龄~6日龄的时候达到最高峰,随后开始逐渐衰减;在6日龄母源抗体达到最高峰时,对低、中等和高水平的母源抗体雏鸡免疫重组乳酸菌r-L.Lactis-VP2-RCK。3个免疫组都能在免疫后1周迅速产生较高的中和抗体。研究表明,重组乳酸菌r-L.Lactis-VP2-RCK免疫雏鸡不受母源抗体干扰,并能产生显著的免疫应答,该研究结果为进一步研制商品化的重组乳酸菌活载体疫苗奠定了基础。 Maternal antibody is an important factor in the early intervention of vaccine immunity.In this paper,we studied the growth and decline of maternal antibodies(IBDV),and when the maternal antibody reached the highest peak.The recombinant Lactococcus lactis rL.Lactis-VP2-RCK was observed whether its immune effect was interfered by maternal antibodies.The results showed that the highest peak of the maternal antibody(IBDV)reached at 3-6 days of age,and then began to gradually decline.When the maternal antibody reached the highest peak at 6 days of age,the chicks in the low,medium and high levels of groups were immunized with the recombinant Lactococcus lactis rL.Lactis-VP2-RCK,and the difference of neutralizing antibody levels between the immunized group and control group was compared by neutralization experiment.The three immunized groups were able to rapidly produce higher neutralizing antibodies one week after immunization,and achieved good immune effects.This study showed that the recombinant Lactococcus lactis r-L.Lactis-VP2-RCK can induce a significant immune response without the maternal antibody interference.This study laid the foundation for the further development of commercial recombinant Lactococcus lactisvaccine.
