基于PPAR信号通路探讨辛温通阳中药对动脉粥样硬化大鼠的保护性机制 被引量:6

Study on the Protective Mechanism of Xinwen Tongyang Chinese Herbal Medicine on Atherosclerosis Rats based on PPAR Signaling Pathway

摘要 目的研究辛温通阳中药对动脉粥样硬化(AS)大鼠的保护作用及机制。方法以1.5F球囊导管造成大鼠主动脉内膜剥脱损伤,并用维生素D3针剂注射加高脂饲料喂养制备AS大鼠模型,随机分为假手术组、模型组、葱白组、桂枝组、辛伐他汀组。造模后第3天开始给药,连续给药4周后取血及胸主动脉损伤段,采用生化检测、油红O染色、酶联免疫吸附试验(ELISA)、免疫印迹试验(Western blotting)检测脂质代谢、炎症因子及相关蛋白表达。结果模型组血清TC、TG、LDL-C浓度明显升高,油红O染色显示血管壁有大量脂质沉积;血清hs-CRP、MMP-9、IL-1、TNF-的浓度明显升高,PPAR、HMG-CoA蛋白表达上调,LXR蛋白表达下调(均P<0.01)。葱白组、桂枝组均在不同程度上降低血清中TC、TG、LDL-C的浓度及hs-CRP、MMP-9、IL-1、TNF-的浓度(P<0.05,P<0.01),血管壁脂质沉积减少,同时下调PPAR、HMGCoA蛋白表达而上调LXR蛋白表达(P<0.05,P<0.01)。结论辛温通阳中药可通过PPAR信号通路调控内源性PPAR,抑制HMG-CoA表达,活化LXR蛋白,发挥抗炎和抗动脉粥样硬化作用。 Objective To investigate the effects and mechanism of Xinwen Tongyang Chinese herbal medicine in atherosclerosis(AS)rats.Methods A rat model of AS was established by 1.5 F balloon catheter and vitamin D3 injection plus high fat diet.They were randomly divided into sham operation group,model group,scallion stalk group,cassia twig group and simvastatin group.On the third day after the model was established,blood samples and thoracic aorta injured segments were taken for 4-week continuous administration.Lipid metabolism,inflammatory factors and related protein expression were detected by biochemical detection,oil red staining,enzyme-linked immunosorbent assay(ELISA)and Western blotting.Results The concentration of TC,TG and LDL-C of model group increased significantly,oil red O staining showed a large amount of lipid deposition in vascular wall;the concentration of hs-CRP,MMP-9,IL-1,TNF-increased significantly,the expression of PPAR,HMG-Co A protein increased,and the expression of LXR protein decreased(all P<0.01).The concentrations of TC,TG,LDL-C,hs-CRP,MMP-9,IL-1 and TNF-were decreased in scallion stalk group,cassia twig group(P<0.05,P<0.01),lipid deposition in vascular wall were decreased,while the expressions of PPAR and HMG-Co A were down-regulated and the expression of LXR protein was up-regulated(P<0.05,P<0.01).Conclusion Xinwen Tongyang Chinese herbal medicine can regulate endogenous PPAR through PPAR signaling pathway,inhibit the expression of HMG-Co A,activate LXR protein,andplay an anti-inflammatory and anti-atherosclerosis role.
