摘要
目的观察活血壮骨汤对膝骨性关节炎动物模型的治疗作用并探讨其机制。方法采用2%木瓜蛋白酶关节腔内注射法建立膝骨性关节炎大鼠模型。以大鼠一般状况、膝关节直径、足趾容积、关节病理切片及国际骨关节炎研究学会(OARSI)评分为指标,观察活血壮骨汤对膝关节炎大鼠的影响。利用ELISA法检测大鼠血清中肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)含量,初步探讨活血壮骨汤治疗膝骨关节炎的作用机制。结果与模型组相比,活血壮骨汤各组均能不同程度地使骨关节大鼠膝关节直径、足趾容积降低、一般状况改善、OARSI评分降低(P<0.05),并能显著降低大鼠血清中TNF-α、IL-1β的含量(P<0.05)。结论活血壮骨汤对膝骨关节炎大鼠具有治疗作用,作用机制可能与其抑制TNF-α、IL-1β分泌有关。
Objective:To explore the therapeutic effect and mechanism of Huoxue Zhuanggu Decoction on knee osteoarthritis animal model. Methods:The rat model of knee osteoarthritis was established by intracavitary injection of 2% papain. To observe the effect of Huoxue Zhuanggu Decoction on knee arthritis rats,the general condition,knee joint diameter,toe volume,joint pathological section and bone joint OARSI score were used as indicators. The contents of IL-1β and TNF-α in serum of rats were detected by ELISA,and the mechanism of the treatment of knee osteoarthritis by Huoxue Zhuanggu Decoction was discussed. Results:Compared with the model group,Huoxue Zhuanggu Decoction could reduce the knee joint diameter,toe volume,general condition and OARSI scores of the osteoarticular rats,and significantly reduce the contents of IL-1β and TNF-α in the serum of the rats(P < 0.05). Conclusion:Huoxue Zhuanggu Decoction has therapeutic effect on knee osteoarthritis rats(P <0.05),and its mechanism may be related to its inhibition of TNF-α and IL-1β secretion.
