摘要
目的:探讨肥胖对乌拉坦诱导的小鼠肺癌模型肿瘤生长的影响及相关信号通路。方法:通过乌拉坦诱导建立C57BL/6J小鼠肺癌模型40只,根据饮食类型及腹腔注射药物不同,分为高脂饮食肺癌模型组(HFD-LC)、高脂饮食对照组(HFD-b)、普通饮食肺癌模型组(CD-LC)和普通饮食对照组(CD-b)。记录各组小鼠体重、肿瘤直径;采用Western blot法检测小鼠肺组织中mTOR信号通路和STAT3信号通路相关蛋白表达;收集肺泡灌洗液,观察计数细胞种类及个数。结果:HFD-b组小鼠的体重增长幅度最快,CD-LC组体重最轻,增长幅度也最小。HFD-LC组与CD-b组小鼠体重及增长幅度均较为接近。实验结束时,HFD-LC组、CD-LC组小鼠的肿瘤平均直径分别为(5.43±0.77)mm、(3.75±0.35)mm,差异具有统计学意义(P<0.001)。与CD-LC组相比,HFD-LC组小鼠肺泡灌洗液中淋巴细胞比例、巨噬细胞比例显著较高。高脂饮食喂养组小鼠的PI3K、p-AKT、AKT、mTOR、p-mTOR、p-STAT3蛋白水平明显高于正常饮食组,经诱导建立肺癌模型后,HFD-LC组所有蛋白表达量的上升幅度明显高于其他组。但STAT3蛋白表达量在两种饮食的肺癌小鼠中无明显差异。结论:肥胖可增强乌拉坦诱导的小鼠肺癌的恶性生物学行为,其机制可能与mTOR和STAT3信号通路活化相关。
Objective:To investigate the effects of obesity on tumor growth and related signaling pathways in urethane-induced lung cancer in mice.Methods:40 lung cancer models of mice were established,which were divided into HFD-LC,HFD-b,CD-LC and CD-b.The body weight and tumor diameter of each group were recorded.The expression of STAT3 signaling pathway and STAT3 signaling pathway related protein in lung tissue was detected by Western blot,and the types and number of cells in alveolar lavage fluid were observed.Results:The weight gain of HFD-b group was the fastest and the CD-LC group was the slowest.Body weight and growth rate of group HFD-LC and group CD-b were close.The average diameter of tumors in HFD-LC group and CD-LC group were(5.43±0.77)mm and(3.75±0.35)mm.Compared with the CD-LC group,the HFD-LC group had significantly higher cell ratio and macrophage ratio in alveolar lavage fluid.The levels of PI3 K protein,p-AKT protein,AKT protein,mTOR protein,p-mTOR,p-STAT3 protein in the high-fat diet group were significantly higher than those in the normal diet group.After the establishment of lung cancer model,the expression of all proteins in the HFD-LC group was significantly higher than that in the other groups.Conclusion:Obesity can enhance the malignant biological behavior of uratan-induced lung cancer in mice.The mechanism may be related to the activation of mTOR signaling pathway and STAT3 signaling pathway.
