异丙酚对小鼠胰腺内分泌细胞的影响及作用机制

摘要 目的:探讨异丙酚在胰岛移植围手术期对小鼠胰腺内分泌细胞的影响及机制。方法:将小鼠胰腺内分泌细胞株Min6分为阳性对照组、常氧培养组、缺氧培养组、常氧加药组、缺氧加药组,加药组以10-6mol/L浓度的异丙酚处理。用免疫印迹试验(Western Blot)、酶联免疫吸附试验(ELISA)法及流式细胞术分别检测细胞缺氧诱导因子-1α(HIF-1α)、血管内皮生长因子(VEGF)表达量及细胞凋亡情况。结果:常氧条件下,Min6细胞加药后HIF-1α、VEGF表达降低,细胞凋率降低(P<0.05);在缺氧条件下,细胞加药后HIF-1α、VEGF降低,细胞凋亡率升高(P<0.05)。结论:异丙酚能够降低常氧条件下小鼠胰腺内分泌细胞的凋亡,其机制可能与抑制HIF-1α/VEGF途径有关,但在缺氧条件下会诱导胰腺内分泌细胞凋亡。 Objective: To investigate the effect and the mechanism of propofol on pancreatic endocrine cells of mice during the perioperative period of islet transplantation.Methods: Pancreatic endocrine cell line Min6 of mice was divided into positive control group,normoxic culture group,hypoxia culture group,normoxia plus drug group,and hypoxia plus drug group.The drug group was treated with propofol at the concentration of 10-6 mol/L.Western Blot,enzyme-linked immunoassay(ELISA),and flow cytometry were used to detect the cell expression level of hypoxia inducible factor-1α(HIF-1α)and vascular endothelial growth factor(VEGF),and cell apoptosis,respectively.Results: Under normoxic condition,the expression level of HIF-1α and VEGF were decreased and the apoptosis rate was decreased after given drugs(P<0.05).Under anoxic condition,the expression level of HIF-1α and VEGF were decreased and the apoptosis rate was increased after given drugs(P<0.05).Conclusion: Propofol can reduce the apoptosis of pancreatic endocrine cells of mice under normoxic condition,and its mechanism may be related to the inhibition of HIF-1α/VEGF,and it can induce pancreatic endocrine cell apoptosis under hypoxia.
