肺脾两虚型慢性阻塞性肺疾病大鼠血清Ghrelin、Obestatin的动态表达 被引量:4

Dynamic Expressions of Ghrelin and Obestatin in the Rats with Chronic Obstructive Pulmonary Disease of Lung and Spleen Deficiency Pattern

摘要 目的:观察肺脾两虚型慢性阻塞性肺疾病(chronic obstructive pulmonary disease,COPD)大鼠血清生长激素释放肽(ghrelin)及肥胖抑制素(obestatin)在不同时间段的动态表达,探究COPD合并营养不良的发病机制。方法:将48只Wistar大鼠随机分为空白对照组、观察组(根据造模时间不同,分为肺脾两虚28、35、42天组)。利用烟熏+气管内滴注脂多糖+灌服冰冷番泻叶浸液造模法建立肺脾两虚型COPD大鼠模型。分别于造模28、35、42天测定大鼠肺功能;HE染色观察大鼠胃组织及肺组织病理变化;酶联免疫吸附法(Elisa法)检测大鼠血清ghrelin、obestatin、瘦素及白细胞介素1(interleukin-1,IL-1)及肿瘤坏死因子α(tumor necrosis factorα,TNF-α)的动态表达水平。结果:与空白对照组比较,观察组各组大鼠最大呼气量(maximal expiratory volume,EV)、呼出50%气量时的流速(exhaled 50%of the flow rate,EF50)、潮气量(tidal volume,TV)、肺动态顺应性(pulmonary dynamic compliance,Cdyn)均降低,肺阻力(lung resistance,LR)增加,差异有统计学意义(P<0.05);与空白对照组比较,观察组各组大鼠ghrelin及obestatin含量降低,血清IL-1、TNF-α及瘦素升高,差异有统计学意义(P<0.01)。观察组各组大鼠肺组织及胃组织病理改变显著。结论:肺脾两虚型COPD大鼠模型ghrelin-obestatin信号调节通路代谢紊乱及炎症因子增高,这可能是COPD合并营养不良的机制之一。 Objective:To explore the pathogenesis of COPD accompanying malnutrition by observing dynamic expressions of ghrelin and obestatin in the rats with chronic obstructive pulmonary disease(COPD)of lung and spleen deficiency pattern at different time periods.Methods:All 48 Wistar rats were randomized into blank control group and the observation group(according to different modeling time,28 days,35 days and 42 days groups of lung and spleen deficiency pattern).Fuming,intratracheal infusion of lipopolysaccharide and drenching of cold Fanxieye(sennae folium)extracts were used to establish COPD rat models of lung and spleen deficiency pattern.Lung functions of the rats were measured 28,35 and 42 days after modeling;pathological changes of stomach and lung tissue of the rats were observed using HE staining;dynamic expressions of TNF-α,IL-1,leptin,ghrelin and obestatin in the serum of the rats were measured using ELISA method.Results:Compared with blank control group,EV(maximal expiratory volume),EF50(exhaled 50%of the flow rate),TV(tidal volume)and pulmonary dynamic compliance(Cdyn)of the rats in the observation group reduced,lung resistance(LR)increased,and the difference had statistical meaning(P<0.05);compared with blank control group,the contents of ghrelin and obestatin reduced,IL-1,TNF-αand leptin rose in the observation group,and the difference showed statistical meaning(P<0.01).Pathological changes of the stomach and lung tissue of the rats in the observation group were striking.Conclusion:The metabolism of ghrelin-obestatin signal regulating pathway in COPD rat models is in disorder and inflammtory factors rise,and they might be one of the mechanisms of COPD accompanying malnutrition.
