摘要
研究普鲁兰酶对蜡质玉米淀粉脱支工艺的影响.以还原力(DE)为考察指标,探讨pH、温度、加酶量和酶解时间对玉米淀粉脱支的影响,采用响应面法对酶解过程中的工艺参数进行优化.研究结果表明,在玉米淀粉脱支工艺单因素试验中最适的酶解条件是:pH 5.0,温度50℃,加酶量7.2 U/g,酶解时间12 h;最佳酶解条件为:pH 5.3,加酶量8.1 U/g,温度50℃.最佳条件下得到的DE为0.57,较单因素试验的DE高,证明此优化条件能较好地提高蜡质玉米淀粉的脱支程度.
To research the effect of pullulanase on waxy corn starch debranching process.The effect of enzyme dosage,pH,temperature and time on the enzymatic hydrolysis of waxy corn starch was investigated with the release rate of reducing sugar(DE)as the index.The response surface methodology was used to optimize the process parameters in the enzymolysis process.Results:The optimum enzymolysis conditions of waxy corn starch pulullanse content of 7.2 U/g,temperature 55℃,enzyme solution pH 5.0,enzymatic hydrolysis time 12 h;Response surface analysis determined the optimal enzymatic conditions were pH 5.3,the amount of enzyme added 8.1 U/g and the temperature 50℃.Under the optimal conditions DE is 0.57,which is higher than that of the single factor test.It is proved that this optimization condition can better improve the debranching degree of waxy corn starch.
