摘要
目的:研究比较包括夏枯草在内的30批唇形科种子挥发油成分,并建立夏枯草种子挥发油(PSV)气相色谱-质谱联用(GC-MS)指纹图谱,测定其抗炎活性。方法:利用GC-MS法分析30批种子挥发油成分,采用“中药色谱指纹图谱计算机相似性评价系统(2004A版)”软件进行模式分析及相似度计算,对共有特征峰成分进行检识;药物干预脂多糖(LPS,0.1μg/mL)建立细胞炎症模型,利用噻唑蓝(MTT)比色法考察不同浓度的PSV对RAW264.7细胞增殖的影响,格里斯试剂法及酶联免疫吸附法检测细胞分泌NO及白介素-6(IL-6)、肿瘤坏死因子α(TNF-α)的水平。结果:30批唇形科种子挥发油的主要成分为不饱和脂肪酸亚油酸、角鲨烯等,其中夏枯草含特有活性成分β-香树脂醇;建立了PSV的GC-MS指纹图谱,鉴定了16个共有特征峰;夏枯草种子挥发油在浓度为23.4~93.8μg/mL时可以显著抑制LPS刺激的RAW264.7细胞分泌NO和TNF-α、IL-6(P<0.05,P<0.01)。结论:PSV含有大量活性成分,本方法重复性好,所建立的指纹图谱为PSV的质量控制提供了有效手段,为PSV的利用提供了重要依据。
Objective:To study and compare the components of volatile oil from 30 species of the Lamiaceae family plants seeds including Prunella vulgaris and establish characteristic fingerprint of volatile oil from Prunella vulgaris seed by gas chromatography-mass spectrometer and to determine the anti-inflammatory activities of the volatile oil.Methods:Chemical components of volatile oil from 30 species of the Lamiaceae family plants seeds were analyzed by GC-MS method,the common model and similarity of the TIC profiles were analyzed by computer aided similarity evaluation system,and the components of common characteristic peaks in chromatograms were identified.Effects of volatile oil from Prunella vulgaris seeds on proliferation of RAW264.7 cells were investigated by MTT assay.Afterwards,RAW264.7 cells were stimulated with LPS,and the production of NO,IL-6 and TNF-αwas detected by the Griess colorimetric method and enzyme-linked immuno sorbent assay.Results:The main components of the volatile oil from 30 species of Labiatae seeds were unsaturated fatty acids.Among them,Prunella vulgaris contained linoleic acid,squalene and beta-amyrin.Fingerprint of volatile oil from Prunella vulgaris seeds was established,and 16 characteristic peaks in GC-MS specific chromatogram of volatile oils from Prunella vulgaris seed were identified and defined.Volatile oil from Prunella vulgaris seeds with the concentration of 23.4~93.8μg/mL could significantly reduce the expression of NO,IL-6 and TNF-α(P<0.05,P<0.01).Conclusion:Volatile oil from Prunella vulgaris seeds contain lots of active components.With a good reproducibility,the established characteristic fingerprints of volatile oil from Prunella vulgaris seed can be used for quality control and provide important basis for application of volatile oils from Prunella vulgaris seed.
