基于“病证-效应-生物样本分析”方法的炮姜温经止血功效物质及归经研究 被引量:10

Study on the related substances and channel tropism of baked ginger on warming meridian and hemostasis based on‘syndrome-efficacy-analysis of biological samples’

摘要 目的:探讨炮姜水煎液对虚寒性出血大鼠模型温经止血功效的物质基础及其体内分布与归经的关系。方法:分别建立脾胃虚寒出血大鼠模型和虚寒性子宫出血大鼠模型,给予炮姜水煎液15g/kg,于末次给药后不同时间点摘眼球取血,脱颈椎处死,立即取心、肝、脾、肺、肾、胃、大肠、小肠等组织。通过对比各器官组织、炮姜药材及标准品HPLC图谱,指认炮姜进入脏器组织的相关物质;并用DAS 3.0软件分析药代动力学参数,探讨炮姜温经止血的功效物质在体内组织分布与归经情况。结果:在两组模型给药大鼠脏器中检测到了姜酮、6-姜酚、6-姜烯酚、8-姜酚、8-姜烯酚、10-姜烯酚6种成分。在脾胃虚寒出血模型大鼠组织中,姜酮AUC0-t较高的为胃、小肠、大肠;6-姜酚AUC0-t较高的为胃、小肠、脾;6-姜烯酚AUC0-t较高的为胃、脾、肝;8-姜酚AUC0-t较高的为脾、心、小肠、肝;8-姜烯酚AUC0-t较高的为胃、心、脾、小肠、肝;10-姜烯酚AUC0-t较高的为小肠、脾、大肠、胃。在虚寒性子宫出血模型大鼠组织中,姜酮AUC0-t较高的为小肠、胃、大肠、肝;6-姜酚AUC0-t较高的为胃、小肠、大肠、子宫、肝;6-姜烯酚AUC0-t较高的为胃、小肠、子宫;8-姜酚AUC0-t较高的为胃、肝、小肠;8-姜烯酚AUC0-t较高的为胃、小肠、心、肝、脾;10-姜烯酚AUC0-t较高的为肝、肺、子宫。结论:炮姜温经止血的功效物质基础为姜酮、6-姜酚、6-姜烯酚、8-姜酚、8-姜烯酚、10-姜烯酚。功效物质在大鼠体内分布与炮姜归肝、脾、胃经的传统认识基本相吻合。 Objective:To investigate the substance basis of decoction of processed baked ginger on the warming meridian and hemostasis effect in rats with deficiency cold hemorrhage,and to explore the relationship between its internal distribution and channel tropism.Methods:Rats with spleen-stomach bleeding due to deficiency and cold and rats with uterine bleeding due to deficiency and cold were established.Decoction of processed baked ginger was given at 15g/kg.The eyeballs were taken out and blood was taken out at different time points after gastric baked ginger decoction,and the cervical vertebrae was removed and executed.The heart,liver,spleen,lung,kidney,stomach,large intestine and small intestine and other tissues were taken out immediately.By comparing the HPLC chromatograms of various organs,medicinal materials and standard substances,the related substances that entered in the organ tissues were identified.The pharmacokinetic parameters were analyzed by DAS 3.0 software,and the pharmacodynamic substances and their distribution in rats were discussed.Results:Six components of gingerone,6-gingerols,6-shogaol,8-gingerol,8-shogaol and 10-shogaol were detected in the viscera of rats in both groups.Among the six substances,in the spleen-stomach deficiency-cold hemorrhage model rats,the stomach,small intestine and large intestine had higher AUC0-t of gingerol,stomach,small intestine and spleen had higher AUC0-t of 6-gingerol,stomach,spleen and liver had higher AUC0-t of 6-shogaol,spleen,heart,small intestine and liver had higher AUC0-t of 8-gingerol,and stomach,heart,spleen,small intestine,and liver had higher AUC0-t of 8-shogaol,small intestine,spleen,large intestine,stomach had higher AUC0-t of 10-shogaol.In uterine bleeding due to deficiency and cold model rats,the AUC0-t of gingerol was higher in small intestine,stomach,large intestine and liver,the AUC0-t of 6-gingerol was higher in stomach,small intestine,large intestine,uterus and liver,the AUC0-t of 6-shogaol was higher in stomach,small intestine and uterus,the AUC
