摘要
获得海葵纯度高、完整性好的高质量RNA,为海葵高通量转录组测序等分子生物学的深入研究提供基础,以南海海葵为研究对象,采用改进TRIzol法提取不同发育时期海葵的总RNA,并利用琼脂糖凝胶电泳、Nano Drop TM 2000分光光度计和Agilent 2100生物分析仪检测其质量。结果表明:改进TRIzol法从不同发育时期的海葵中提取的RNA电泳条带清晰、海葵-大与海葵-中的RNA完整性好,海葵-小RNA完整性不合格;海葵-大、海葵-中和海葵-小的纯度(OD260/280)分别为1.981、1.983和2.071,浓度分别为335ng/μL、357ng/μL和232ng/μL。改进TRIzol法可有效从海葵中提取高质量RNA。
Obtaining high-quality RNA with high purity and wonderful integrity of sea anemones would provide a basis for in-depth studies of molecular biology such as high-throughput transcriptome sequencing of sea anemones,in this paper the authors used South China Sea anemones as the research object,and used improved TRIzol method to extract total RNA of sea anemone in different developmental stages,and the quality was tested by agarose gel electrophoresis,Nano Drop TM 2000 spectrophotometer and Agilent 2100 bioanalyzer.Results:The improved TRIzol method obtained clear electrophoretic bands of RNA extracted from anemones in different developmental stages,the integrity of RNA in large and medium anemones was good,and the RNA integrity of small anemones was unqualified;The purity(OD260/280)of large,medium and small sea anemones was respectively 1.981,1.983 and 2.071,and their concentrations were 335 ng/μL,357 ng/μL and 232 ng/μL,respectively.The improved TRIzol method can effectively extract high-quality RNA from sea anemones.
