摘要
目的 建立一种高效快速提取大鼠心肌总RNA的方法 ,为心肌组织mRNA的检测提供条件。方法 利用高浓度强烈变性剂 (异硫氰酸胍 )使细胞结构迅速破坏 ,并直接通过酚等有机溶剂处理、离心 ,使RNA与细胞DNA、蛋白质、细胞残片等分离 ,迅速得到总RNA。结果 得到高纯度、未被降解的总RNA。结论 此法简洁高效 ,耗时短 ,完全可用于临床分析工作。
Objective To establish an efficient and quick method of total RNA of cardiac muscle in rats, and to provide conditions as the basis is of analysis of mRNA in cardiac muscle. Methods To make the cell construction be broken quickly by making use of the strong denaturant (guanidium thiocyanate),and to be dealt with directly by adding pheno and organic solution,and the RNA was separated to DNA, protein and cell's relic etc. by spinning, finally to get the total RNA quickly Results Getting the high-purity and undegraded RNA. Conclusion This is a kind of rapid and efficient purification method. Not only does it shorten the experiment's time but also improve the quality. The method can be completely used for the clinical analysis work.
