早小洋菊和射阳大白菊对脂多糖诱导RAW264.7细胞炎症反应影响的差异 被引量:1

摘要 目的评价早小洋菊和射阳大白菊抗炎作用及差异。方法将RAW264.7细胞按5×10^4个/孔接种于6孔细胞培养板中培养。空白组不做任何处理正常培养24h;模型组加入脂多糖(LPS)500ng/ml诱导RAW264.7细胞建立炎症模型;早小洋菊和射阳大白菊高、低剂量组分别加入浓度为125、62.5μg/ml相应菊花醇提取物1ml/孔,作用2h后再加入LPS溶液500ng/ml刺激24h。每组设5个复孔。收获细胞上清液检测一氧化氮(NO)浓度,RT-PCR法检测细胞中肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)、白细胞介素1β(IL-1β)、诱导性一氧化氮合酶(iNOS)、环氧化酶2(COX-2)mRNA表达水平。结果与空白组比较,模型组上清液中NO水平和细胞中TNF-α、IL-6、IL-1β、iNOS、COX-2 mRNA表达均上调(P<0.01)。与模型组比较,除早小洋菊高剂量组细胞中IL-1βmRNA表达外,其余各菊花组上清液中NO水平和细胞中TNF-α、IL-6、IL-1β、iNOS、COX-2 mRNA表达均下调(P<0.01)。早小洋菊高剂量组细胞中iNOS mRNA表达较射阳大白菊高剂量组降低,TNF-α、IL-1βmRNA表达升高(P<0.01);早小洋菊低剂量组细胞中IL-6、IL-1β、COX-2 mRNA表达较射阳大白菊低剂量组降低,TNF-αmRNA表达升高(P<0.01)。结论早小洋菊和射阳大白菊均具有较好的抗炎活性,对于炎症因子iNOS、COX-2、IL-6的抑制作用早小洋菊要强于射阳大白菊,对于TNF-α和IL-1β的抑制作用射阳大白菊强于早小洋菊。 Objective To evaluate anti-inflammatory effect and difference of Early Chrysanthemum and Sheyang White Chrysanthemum. Methods RAW264.7 cells were inoculated into 6-hole cell culture plates at 5 × 10^4 cells/hole. Blank group was cultured for 24 h without any treatment;model group was added with 500 ng/ml lipopolysaccharide(LPS) to induce RAW264.7 cells to establish inflammation models;high and low dose groups of Early Chrysanthemum and Sheyang White Chrysanthemum were added with 125 and 62.5μg/ml corresponding chrysanthemum alcohol extracts for 1 ml/hole, respectively, and 2 h after the treatment, they were stimulated by 500 ng/ml LPS solution for 24 h. Five parallel holes were set in each group. Supernatant of the cells was collected to detect the concentration of nitric oxide(NO), and tumor necrosis factor α(TNF-α), interleukin 6(IL-6), interleukin 1β(IL-1β), inducible nitric oxide synthase(iNOS) and cyclooxygenase 2(COX-2) mRNA expression levels were detected by RT-PCR. Results Compared with the blank group, the level of NO in the supernatant and the expressions of TNF-α, IL-6, IL-1β, iNOS and COX-2 mRNA in the cells of the model group were up-regulated(P<0.01). Compared with the model group, except for the expression of IL-1β mRNA in the cells of the high dose group of Early Chrysanthemum, the levels of NO in the supernatant and the cell TNF-α, IL-6, IL-1β, iNOS and COX-2 mRNA expressions in the other medicated groups were down-regulated(P<0.01). The expression of cell iNOS mRNA in the high dose group of Early Chrysanthemum was lower than that in the high dose group of Sheyang White Chrysanthemum, and the expressions of TNF-α and IL-1β mRNA were increased(P<0.01). The expressions of cell IL-6, IL-1β and COX-2 mRNA in the low dose group of Early Chrysanthemum were lower than those in the low dose group of Sheyang White Chrysanthemum, and the expression of TNF-α mRNA was increased(P<0.01).
