Optimization of Alcohol-water Double Extraction Process of Gange Hugan Capsule by Box-Behnken Response Surface Method

摘要 目的优选甘葛护肝胶囊的提取工艺。方法依据单因素试验结果,采用Box-Behnken设计,以木香烃内酯(Costunolide,CLD)、去氢木香内酯(Dehydrocostuslactione,DYDL)的转移率和醇提浸膏得率为醇提综合指标(Y1),优选最佳醇提工艺。以葛根素的转移率和水提浸膏得率为水提综合指标(Y2)优选最佳水提工艺,并对工艺进行验证。结果醇提最佳条件为乙醇体积分数85.77%,提取时间107.56 min,提取次数2.61次,料液比1∶15.87(g:ml),验证试验中Y1均值为101.93分(RSD=2.89%,n=3),与预测值104.71分相比较,相对误差为2.65%。水提最佳条件为提取次数2.82次,料液比1∶10.97(g:ml),提取时间100.03 min,验证试验中Y2均值为96.42分(RSD=2.42%,n=3),与预测值98.01分相比较,相对误差为1.62%。结论所优选甘葛护肝胶囊提取工艺稳定可行。 Objective To optimize the optimal extraction process of Gange Hugan Capsule.Methods In the process of optimizing the optimal ethanol extraction process,according to the single factor test and the Box-Behnken response surface method,the comprehensive index of the ethanol extraction process(Y1)includes the transfer rate of the costunolide and dehydrocostus,the amount of ethanol extract into the extract.In the process of optimizing the optimal water extraction process,the transfer rate of puerarin and the amount of water extract to the extract are comprehensive indicators of water extraction(Y2),and the process is verified.Results The optimum conditions for ethanol extraction are ethanol volume fraction of 85.77%,extraction time of 107.56 min,extraction times of 2.61 times,solid-liquid ratio of 1:15.87(g:ml),and the average value of Y1 in the verification test is 101.93 points(RSD=2.89%,n=3),compared with the predicted value of 104.71,the relative error is 2.65%;The optimum conditions for water extraction are extraction times of 2.82 times,solid-liquid ratio of 1:10.97(g:ml),extraction time of 100.03 min,and the average value of Y2 in the verification test is 96.42 points(RSD=2.42%,n=3),compared with the predicted value of 98.01,the relative error is 1.62%.Conclusion The preferred extraction process of Gange Hugan Capsule is stable and feasible.
