木霉菌Strain T1122分子鉴定及其降氮除磷效应研究 被引量:1

摘要 研究中对一株木霉菌(Strain T1122)进行了分子鉴定,并评估其在污水处理中的降氮除磷效应.菌落形态观察、序列比对及系统进化分析等结果表明,分离到的菌株隶属于木霉属中的绿色木霉(Trichoderma virens).将该菌株接种至含铜绿微囊藻的高氮磷污水中进行培养,分别设置灭活和未灭活铜绿微囊藻液作为对照组(CK1、CK2)以及灭活铜绿微囊藻液加Strain T1122和未灭活铜绿微囊藻加Strain T1122作为实验处理组(处理1和处理2).研究结果显示:处理1氮和磷含量持续快速下降,培养相同天数时,其氮磷含量均显著低于CK1.处理2在培养第3 d,其氮含量显著低于CK1,显著高于CK2;磷含量均显著低于CK1和CK2.另外研究发现,菌体培养3 d处理2溶液中铜绿微囊藻细胞密度仅为CK2的3.56%,表明该菌对铜绿微囊藻有极强的吸附作用.本研究发现该种木霉能够吸附铜绿微囊菌且能有效降低污水氮磷含量,这为绿色木霉在污水处理中的应用提供了依据. In this study,Strain T1122(a Trichoderma strain)was identified,and its effect of nitrogen and phosphorus reduction in wastewater treatment was evaluated.The results of colony morphology,sequencing and phylogenetic tree analysis showed that the colony we isolated belonged to Trichoderma virens.The strain was inoculated into the wastewater containing Microcystis aeruginosa,nitrogen and phosphorus for cultivation,and four treatments were set up,including the control of sterile Microcystis aeruginosa solution(CK1),the control of Microcystis aeruginosa solution(CK2),the treatment of Microcystis aeruginosa solution with Trichoderma(treatment 1)and the treatment of Microcystis aeruginosa with Trichoderma(treatment 2).The content of nitrogen and phosphorus in treatment 1 decreased continuously and rapidly.The content of nitrogen and phosphorus in treatment 1 was significantly lower than that of CK1 in the same days of culture.On the third day of culture,nitrogen content of treatment 2 was significantly lower than CK1,but significantly higher than CK2;phosphorus content was significantly lower than CK1 and CK2.Trichoderma had strong adsorption on Microcystis aeruginosa.The cell density of Microcystis aeruginosa in treatment 2 was only 3.56%of that in CK2 on the third day of culture.This study provides a basis for the application of Trichoderma viride in wastewater treatment.
