摘要
目的探讨肾透明细胞癌(ccRCC)患者外周血微小RNA-223(miR-223)的表达及其与临床病理参数的关系。方法收集40例ccRCC患者术前及术后7 d外周血标本为实验组,收集80例健康人群外周血标本为对照组。采用实时定量聚合酶链式反应(RT-PCR)仪器检测miR-223在实验组与对照组中的表达水平,采用受试者工作特征(ROC)曲线评价外周血中的miR-223表达水平诊断ccRCC的价值。分析ccRCC患者外周血miR-223表达水平与患者病理特征的关系。结果本研究结果显示,ccRCC患者术前外周血中miR-223的表达量为(0.568±0.047),显著高于健康人群的(0.231±0.037)(P<0.05);ccRCC患者术后7 d外周血中miR-223的表达量为(0.324±0.041),较术前外周血中miR-223表达量降低,但仍显著高于健康人群(P<0.05)。同时,miR-223在外周血中的表达与肿瘤TNM分期、肿瘤大小、肿瘤分化程度有显著相关性(P<0.05),与年龄、性别、有无淋巴结转移无显著相关性(P>0.05)。外周血中miR-223的表达在区分ccRCC患者和健康人群中具有较高诊断价值,其ROC曲线下面积为0.893。结论在ccRCC患者外周血中,miR-223呈高表达,且与肿瘤的临床病理特征有一定相关性,对患者术后的复查及预后判断亦有一定的价值。
Objective To investigate the expression of microRNA-223(miR-223)in peripheral blood of patients with clear cell renal cell carcinoma(ccRCC)and its relationship with clinical pathological characteristics.Methods The peripheral blood samples of 40 patients with ccRCC before operation and 7 days after operation were collected as experimental group,and the peripheral blood samples of 80 healthy people were collected as control group.Real-time quantitative PCR(RTPCR)was used to detect the expression level of miR-223 in the experimental group and the control group.The ROC curve was used to evaluate the value of peripheral miR-223 level in diagnosis of ccRCC.The relationship between the expression of miR-223 in peripheral blood and the pathological characteristics was analyzed in patients with ccRCC.Results The results showed that the expression of miR-223 in the peripheral blood of patients with ccRCC before operation was(0.568±0.047),which was significantly higher than(0.231±0.037)in the healthy people(P<0.05).The expression of miR-223 in the peripheral blood of patients with ccRCC at 7 days after operation was(0.324±0.041),which was lower than that before operation,but was still significantly higher than that in the healthy people(P<0.05).The expression of miR-223 in peripheral blood was significantly correlated with TNM stage,tumor size and tumor differentiation(P<0.05),but showed no correlations with age,gender and lymph node metastasis(P>0.05).The expression of miR-223 in peripheral blood showed a high diagnostic value in distinguishing ccRCC cases from healthy people,and area under ROC curve was 0.893.Conclusion The miR-223 is highly expressed in the peripheral blood of patients with ccRCC,which is related to the clinicopathological features of the tumor,and has a certain value for the postoperative re-examination and judgment of prognosis.
