摘要
目的:探讨圣草酚(EDT)干预实验性结肠炎小鼠对Nrf2/HO-1通路的影响。方法:将32只雄性C57BL/6小鼠随机分为空白对照组、DSS模型组、DSS+EDT低剂量组(EDT 20 mg/kg)、DSS+EDT高剂量组(EDT 40 mg/kg)4组,每组8只,小鼠自由饮用3%DSS溶液制成溃疡性结肠炎模型,并同时予以圣草酚腹腔注射7 d。对小鼠进行疾病活动指数(DAI)评分;比较各组小鼠结肠长度;HE染色观察结肠病理学变化;检测结肠组织MDA含量、MPO和SOD活性;Real-time PCR法检测各组小鼠结肠组织Nrf2、HO-1、NOQ1、TNF-α、IL-6 mRNA表达情况;Western blot检测各组小鼠结肠组织中Nrf2、HO-1、NOQ1变化情况。结果:与DSS组比较,空白组、圣草酚低、高浓度治疗组DAI评分、结肠长度及病理学变化明显改善,差异有统计学意义(P<0.05)。空白组、圣草酚低、高浓度治疗组炎症因子TNF-α、IL-6水平,MPO活性及MDA含量显著低于模型组,SOD活性显著高于模型组,差异有统计学意义(P<0.05)。与DSS组相比,圣草酚低、高浓度组显著增加Nrf2、H0-1、NOQ1表达(P<0.01)。结论:圣草酚对小鼠结肠炎可以起到缓解作用,可能与激活Nrf2/HO-1通路发挥抗氧化、抗炎作用有关。
Objective:To investigate the effect of eriodictyol(EDT)on the Nrf2/HO-1 pathway in experimental colitis mice.Methods:Thirty two male C57BL/6 mice were randomly divided into control group,DSS model group,DSS+EDT low dose group(EDT 20 mg/kg),DSS+EDT high-dose group(EDT 40 mg/kg),8 mice in each group.Ulcerative colitis mice model was induced by 3%DSS,and eriodictyol was injected intraperitoneally for 7 days.The disease activity index(DAI)scores were examined;the colon length of each group was compared;the pathological changes of colon were observed by HE staining;the content of MDA,the MPO and SOD levels were detected;the mRNA expressions of Nrf2,HO-1,NOQ1,TNF-αand IL-6 of each group were detected by Real-time PCR method.The changes of Nrf2,HO-1 and NOQ1 in colon tissue of each group were detected by Western blot.Results:Compared with the DSS group,DAI,colon length and pathological changes were significantly improved in the blank group,the low-dose group and the high-dose eriodictyol groups,the difference was statistically significant(P<0.05).In the low-dose group,the high-dose eriodictyol treatment and blank groups,the levels of inflammatory factors TNF-αand IL-6,the activity of MPO and the content of MDA were significantly lower than the model group,the activity of SOD was significantly higher than the model group,and the difference was statistically significant(P<0.05).Compared with the DSS group,the low-dose,high-dose eriodictyol group enhanced the expression of Nrf2,HO-1 and NOQ1 in the colon(P<0.01).Conclusion:The eriodictyol can play a protective role in colitis mice,which may be related to the anti-inflammatory and antioxidant effect of enhancing the Nrf2/HO-1 pathway.
