马氏珠母贝黏液中天然牛磺酸分离纯化及其结构表征 被引量:4

Separation, Purification and Structural Characterization of Natural Taurine from the Mucus of Pinctada Martensii

摘要 以马氏珠母贝黏液为原料,等电点沉淀法脱除黏液中蛋白,经强酸性阳离子交换树脂、乙醇结晶获得牛磺酸晶体,同时采用高效液相色谱法测定其纯度,傅利叶红外光谱及扫描电镜对其结构进行表征。结果显示:提取的天然牛磺酸呈白色粉末状,提取率为0.68 mg/mL;提取的天然牛磺酸晶体红外光谱图与合成牛磺酸标准品基本一致;扫描电镜观察可见提取的天然牛磺酸晶体呈针状,外形棱角分明,表面粗糙,附着较多细小晶粒;高效液相色谱法测定提取的天然牛磺酸晶体纯度为85.2%。 In this paper,the mucus of Pinctada martensii was used as raw materials,the protein in the mucus was removed by isoelectric precipitation,and the crystal of taurine was obtained by strong acid cation exchange resin and ethanol crystallization.Meanwhile,the purity was determined by high performance liquid chromatography and the structure was characterized by infrared spectroscopy and scanning electron microscopy.The results showed that the extracted natural taurine was white powder with an extraction rate of 0.68 mg/mL,the infrared spectrum of the extracted natural taurine crystal was basically consistent with the synthetic taurine standard,the extracted natural cattle were observed by scanning electron microscopy.The taurine crystal has a needle shape,a sharp outline,a rough surface,and a large number of small crystal grains.The purity of the extracted natural taurine crystal is 85.2% by high performance liquid chromatography.
