基于Oncomine和TCGA数据库分析CHRNA5在肺鳞癌中的表达及临床意义

摘要 为了分析烟碱型胆碱能受体5(cholinergic receptor nicotinic alpha 5 subunit,CHRNA5)在肺鳞癌中的表达及其临床意义,利用Oncomine数据库信息分析CHRNA5基因在肿瘤及肺鳞癌组织中的表达情况;利用TCGA数据库信息分析CHRNA5基因在肺鳞癌患者中的表达及与临床参数的相关性;利用免疫组化方法检测52例临床肺鳞癌组织样本和52例癌旁组织样本中CHRNA5编码蛋白α5-nAchR的表达情况,探讨α5-nAchR表达与患者临床病理特征的关系。结果表明:Oncomine数据库中共有402项关于CHRNA5在不同肿瘤表达水平的相关研究,CHRNA5表达存在统计学差异有27项,其中有4项研究显示,CHRNA5在肺癌组织与正常组织中的表达有显著差异,且2项表明CHRNA5在肺鳞癌中的表达量显著高于正常组织.TCGA数据库结果显示,α5-nAchR表达水平与肺鳞癌患者的年龄、性别、吸烟史以及吸烟量密切相关.52例肺鳞癌组织样本免疫组织化学结果显示,α5-nAchR蛋白在肺鳞癌组织中的表达明显高于癌旁组织,且在肺鳞癌样本中α5-nAchR的表达和患者吸烟史密切相关.本研究表明CHRNA5基因在吸烟相关性肺鳞癌发生发展中可能发挥重要作用,CHRNA5有望成为肺癌治疗的靶点之一,为进一步阐明其在吸烟相关性肿瘤发生中的作用提供理论基础. To analyze the expression of cholinergic receptor nicotinic alpha 5 subunit(CHRNA5)and its clinical significance in lung squamous cell carcinoma(LUSC),we analyzed the expression of CHRNA5 in lung cancer tissues by Oncomine database.The correlation between the expression of CHRNA5 in LUSC and clinical parameters were analyzed by TCGA.Immunohistochemical detection was performed in 52 LUSC tissues and 52 para-carcionma tissues.The relationship betweenα5-nAchR expression and clinicopathological parameters was analyzed.The results show that 402 studies on the expression levels of CHRNA5 in different tumors were collected from the Oncomine database,in which there were 27 studies significant differences in CHRNA5 expression and 4 studies showed significant differences in CHRNA5 expression between lung cancer tissues and para-carcionma tissues,including 2 LUSC studies.α5-nAchR expression levels were significantly associated with age,gender,smoking history,and smoking in LUSC tissues by TCGA.Immunohistochemical staining results showed that the expression ofα5-nAchR in LUSC patients was significantly higher than that in para-carcionma tissues.The higher expression ofα5-nAchR was closely related with the smoking history of patients in LUSC tissues.In conclusion,CHRNA5 might play an important role in the development of smoking-related LUSC,which will be expected to become one of potential targets of lung cancer treatment.
