摘要
目的:建立黄花柳花中芦丁、木犀草素-7-О-β-D-吡喃葡萄糖苷、芹菜素-7-O-β-D-吡喃葡萄糖苷、木犀草素、芹菜素的HPLC分析方法。方法:采用Agilent TC-C18色谱柱(4.6mm×250mm,5μm),以乙腈-0.2%磷酸溶液梯度洗脱,检测波长340nm,流速0.8mL/min,柱温30℃。结果:芦丁、木犀草素-7-О-β-D-吡喃葡萄糖苷、芹菜素-7-O-β-D-吡喃葡萄糖苷、木犀草素、芹菜素的线性范围分别为8.11~121.68μg/mL(r=0.9994),2.92~43.80μg/mL(r=0.9995),4.13~61.92μg/mL(r=0.9995),2.04~30.60μg/mL(r=0.9995),2.05~30.78μg/mL(r=0.9996);平均回收率(n=9)分别为100.25%(RSD=1.60%),98.24%(RSD=1.23%),101.57%(RSD=1.25%),98.45%(RSD=1.42%),101.30%(RSD=1.17%)。结论:本方法操作简便,快速准确,可以用于黄花柳花中黄酮类成分的定量分析。
Objective:To establish a high performance liquid chromatography (HPLC) method for the determination of Rutin,luteolin-7-O-β-D-pyranoside,apigenin-7-O-β-D-pyranoside,luteolin and apigenin in flower of Salix caprea L..Methods:Agilent TC-C18 column (4.6mm×250mm,5μm) was used to elucidate with acetonitrile-0.2% phosphoric acid solution.The detection wavelength was 340nm,the flow rate was 0.8mL/min,and the column temperature was 30℃.Results:The linear ranges of rutin,luteolin-7-O-β-D-pyranoside,apigen-7-O-β-D-pyrrolidine,luteolin and apigenin were 8.11~121.68μg/mL (r=0.9994),2.92~43.80μg/mL (r=0.9995),4.13~61.92μg/mL(r=0.9995),2.04~30.60μg/mL (r=0.9995),and 2.05~30.78μg/mL(r=0.9996) respectively.The average recovery (n=9) was 100.25% (RSD=1.60%),98.24% (RSD=1.23%),101.57% (RSD=1.25%),98.45% (RSD=1.42%) and 101.30% (RSD=1.17%) respectively.Conclusion:The method established in this article is simple,rapid and accurate,and can be used for quantitative analysis of flavonoids in flower of Salix caprea L..
