摘要
目的研究羟基红花黄色素A(hydroxysafflor yellow A,HSYA)体外模拟胃肠道系统中的稳定性,口服后小鼠组织中的分布,以及HSYA跨小肠上皮细胞膜运输能力。方法体外模拟胃肠道消化实验采用紫外分光光度法检测体外模拟胃肠道消化分别为1和2h后的HSYA的全光谱特性以及高效液相色谱法(high performance liquid chromatography,HPLC)测定HSYA的含量,动物实验包括3种不同摄入方式:腹腔注射、灌胃及长期饲喂。腹腔注射和灌胃给药方式分别有两个时间点:1和2 h,剂量为0.01 mL/g;长期饲喂方式为含有2.5%红花小花饲喂10周,采集小鼠组织,样品处理采用6%高氯酸和甲醇沉淀蛋白法,利用HPLC测定样本中HSYA的含量。HSYA与小肠上皮细胞共培养不同时间后,吸取培养液,把细胞裂解后,通过HPLC测定培养液和裂解后的HSYA含量。结果体外消化实验结果表明,胃、肠道消化后的HSYA结构未被破坏,HSYA在胃肠道的稳定性较好;腹腔注射红花水提物后HSYA在小鼠血清、肝脏、肾、肺中的含量随着时间延长而下降,只有肌肉中HSYA的含量随着时间的延长而升高,而脾、睾丸、脑、心中均未检测到HSYA。灌胃和长期饲喂红花水提物后小鼠组织和血清中未检测到HSYA,表明HSYA无法经口服被吸收入血。通过细胞实验进一步证明了HSYA无法透过小肠上皮细胞而实现跨膜转运。结论本研究证明了HSYA在胃肠道中的稳定性较好,但口服后不能跨过肠道屏障被吸收入血;HSYA以注射方式入血后在组织中的分布存在一定差异。
Objective To study the stability of hydroxysafflor yellow A(HSYA)in simulated gastrointestinal system in vitro,the distribution in mouse tissues after oral administration,and the transport ability of HSYA across intestinal epithelial cell membrane.Methods The ultraviolet spectrophotometry was used to detect the full-spectrum characteristics of HSYA after in vitro simulated gastrointestinal digestion for 1 and 2 h,and the content of HSYA was determined by high performance liquid chromatography(HPLC)in In vitro simulated gastrointestinal digestion experiment.Animal experiments include three different intake methods:intraperitoneal injection,gavage and long-term feeding.Intraperitoneal injection and intragastric administration had 2 time points of 1 and 2 h,respectively,and the dose was 0.01 mL/g.The long-term feeding method contained 2.5%safflower florets for 10 weeks,and the mouse tissues were collected.The sample was treated with 6%perchloric acid and methanol precipitation protein method,and the content of HSYA in the sample was quantified by HPLC.In cell experiments,HSYA was co-cultured with intestinal epithelial cells for different time,and the culture medium was taken.After the cells were lysed,the content of HSYA in the culture medium and after lysis was determined by HPLC.Results The results of in vitro digestion experiments showed that the structure of HSYA was not destroyed after digestion in the stomach and intestine,and the stability of HSYA in the gastrointestinal tract was good.After acute intraperitoneal injection of safflower aqueous extract,the content of HSYA in serum,liver,kidney and lung of mice decreased with time,only the content of HSYA in muscle increased with time,while HSYA was not detected in spleen,testis,brain and heart.HSYA was not detected in the tissues and serum of mice after acute gavage and long-term feeding of safflower aqueous extract,indicating that HSYA could not be absorbed into the blood by oral administration.Through cell experiments,it was further proved that HSYA could no
