摘要
目的探讨浆细胞样树突状细胞(pDC)和TLR7/9通路在寻常型银屑病发病机制中的作用。方法选取19例寻常型银屑病患者的皮损组织和皮损旁正常组织为研究材料,采用免疫组化方法检测pDC的特异性表面标志分子CD2AP、CD123和TLR7、TLR9及其信号通路中相关蛋白分子白细胞介素1受体相关激酶1(IRAK-1)、干扰素调节因子7(IRF7)的表达情况并进行对比分析。结果皮损组织中CD2AP+pDC和CD123+pDC的阳性率均明显高于皮损旁正常组织(均P<0.05),在皮损组织表皮层和真皮层均有分布,但以真皮层居多。皮损组织中TLR7、TLR9、IRAK-1、IRF7在真皮层的表达较为明显,表达阳性率均高于皮损旁正常组织(均P<0.05);TLR9、IRAK-1、IRF7在表皮层均有少量表达,表达阳性率高于皮损旁正常组织(均P<0.05),而TLR7在表皮层的表达阳性率与皮损旁正常组织相比无统计学差异(P>0.05)。结论CD2AP+pDC、CD123+pDC在寻常型银屑病皮损组织中异常分布,可能与TLR7/9通路的激活及其下游途径有关,TLR信号异常活化对寻常型银屑病的发生发展起一定作用。
Objective To investigate the distribution of plasma cell-like dendritic cells(pDC)and the expression of TLR7/9 pathway related molecules in psoriasis vulgaris.Methods Tissue samples from the skin lesion and adjacent normal skin were taken from 19 patients with psoriasis vulgaris.The expression of pDC specific surface markers CD2AP and CD123,and the TLR7/9 pathway related molecules TLR7,TLR9,IRAK-1 and RF7 in lesion tissue and normal skin samples was detected with immunohistochemical methods.Results The positive rates of CD2AP+pDC and CD123+pDC in skin lesions were significantly higher than those in the adjacent normal tissues(both P<0.05).The pDCs were distributed in the epidermal and dermal layers of the lesions,but most of them were in the dermis.The expression of TLR7,TLR9,IRAK-1 and IRF7 in the dermis of the skin lesions was significantly higher than that in the dermis of the normal skin tissues(all P<0.05).The expression of TLR9,IRAK-1,and IRF7 in epidermal layer of the skin lesions was higher than that of adjacent normal tissues(all P<0.05),while there was no significant difference in expression of TLR7 in the epidermal layer between skin lesions and the normal skin tissues(P>0.05).Conclusion The abnormal distribution of CD2AP+pDC and CD123+pDC in psoriatic vulgaris tissues is closely related to its pathogenesis,which may be associated with the activation of the TLR7/9 pathway and its downstream signaling.
