摘要
目的通过检测子痫前期孕妇胎盘组织中Th22、CD4+CD25+CD127low Treg细胞转录因子AHR、Foxp3及细胞因子IL-22、IL-10的表达,探讨其在子痫前期中的作用。方法选取35例明确诊断的子痫前期孕妇为研究组,同时选取正常妊娠孕妇35例为对照组,采用实时荧光定量PCR检测两组孕妇胎盘组织中Th22、CD4+CD25+CD127low Treg细胞转录因子AHR mRNA、Foxp3 mRNA的表达情况;采用免疫组化检测细胞因子IL-22、IL-10的表达。结果AHR mRNA在研究组和对照组的表达量分别为(1.85±0.47)和(0.89±0.26),Foxp3 mRNA在两组的表达量分别为(0.76±0.23)和(1.31±0.42),差异均具有统计学意义;IL-22在研究组和对照组的阳性表达率分别为68.57%和40.00%,IL-10在两组的表达率为34.29%和60.00%,差异也具有统计学意义。结论Th22、Treg细胞转录因子及细胞因子的异常表达,可能参与了子痫前期的发生。
Objective To investigate the expression of Th22,CD4+CD25+CD127low Treg cell transcription factors AHR,Foxp3,and cytokines IL-22 and IL-10 in the placenta of preeclampsia pregnant women,and to explore their role in preeclampsia.Methods Thirty-five definite pregnant women with preeclampsia were selected as the study group,while 35 normal pregnant women were selected as the control group.The Th22,CD4+CD25+CD127low Treg cell transcription factor AHR mRNA and Foxp3 mRNA expressions in placental tissues of two groups of pregnant women were detected by real-time fluorescence quantitative PCR.The cytokine IL-22 and IL-10 expressions were detected by immunohistochemical method.Results The expression levels of AHR mRNA in the study group and control group were(1.85±0.47)and(0.89±0.26),respectively.The expression levels of Foxp3 mRNA in the two groups were(0.76±0.23)and(1.31±0.42),and the differences were statistically significant.The positive expression rates of IL-22 in the study group and control group were 68.57%and 40.00%,respectively.The expression rates of IL-10 in the two groups were 34.29%and 60.00%,and the differences were also statistically significant.Conclusion The abnormal expression of Th22 and Treg cell transcription factors and cytokines may be involved in the occurrence of preeclampsia.
