131I治疗老年分化型甲状腺癌患者的效果分析及对胞外信号调节激酶通路蛋白的调控作用研究 被引量:5

摘要 目的分析131I治疗老年分化型甲状腺癌患者的效果及对胞外信号调节激酶(ERK)通路蛋白的调控作用。方法对180例老年分化型甲状腺癌患者的临床资料进行研究,分析患者经131I治疗后血清中血细胞及肝肾功能变化,并检测患者血清中ERK通路蛋白水平变化。结果细胞数和血小板数均低于治疗前,差异均有统计学意义(P﹤0.05),而治疗前后红细胞数、淋巴细胞数、谷丙转氨酶(ALT)、谷草转氨酶(AST)、血尿素氮(BUN)、碱性磷酸酶(ALP)及肌浆网钙离子ATP酶2a(SERCA2a)水平比较,差异均无统计学意义(P﹥0.05)。血清中ERK信号通路蛋白B细胞淋巴瘤/白血病-2相关X蛋白(BAX)、p53、p73蛋白水平均升高而Ras、Raf、胞外信号调节蛋白激酶(MEK)、ERK1/2、B细胞淋巴瘤/白血病-2(Bcl-2)、高迁移率族蛋白B1(HMGB1)及增殖细胞核抗原(PCNA)蛋白水平均降低,差异均有统计学意义(P﹤0.05)。结论131I治疗老年分化型甲状腺癌患者效果与其调控患者体内ERK信号通路有关,且不影响患者的肝肾功能。 Objective The research aims to investigate the effect of 131I on differentiated thyroid carcinoma in elderly patients and its regulative effect on extracellular signal-regulated kinase(ERK)signaling pathway-related proteins.Method The clinical data of 180 elderly patients with differentiated thyroid carcinoma were retrospectively analyzed,the change of hematology,liver and renal function after 131I therapy were analyzed,besides,the change of ERK signaling pathway related proteins in serum of patients were also detected.Result After 131I treatment in these elderly patients with differentiated thyroid cancer,the white blood cell count and platelet count in serum were significantly decreased(P<0.05),though the red blood cell count,lymphocyte count,alanine aminotransferase(ALT),aspartate aminotransferase(AST),blood urea nitrogen(BUN),alkaline phosphatase(ALP)and sarcoplasmic reticulum calcium ATPase 2a(SERCA2a)after treatment remained in similaritywith those before(P>0.05).The ERK signaling pathway-related proteins Bcell lymphoma/leukemia-2 associated X protein(BAX),p53,p73 were increased greatly,while the Ras,Raf,mitogen-activated protein kinase(MEK),ERK1/2,B cell lymphoma/leukemia-2(Bcl-2),high mobility group box 1(HMGB1),and proliferating cell nuclear antigen(PCNA)were decreased significantly after the treatment of 131I(P<0.05).Conclusion 131I therapy is effective for the treatment of differentiated thyroid carcinoma,and the effect is notably related to the regulation of ERK signaling pathway,without compromising the liver and renal function of patients.
