摘要
目的探讨结肠癌组织中Musashi 1蛋白和微小RNA-132(miRNA-132)的表达情况及与患者临床特征的关系。方法选择80例结肠癌患者的结肠癌组织标本及其癌旁组织标本,分别采用实时荧光定量聚合酶链反应和免疫组织化学染色方法检测两种组织中miRNA-132和Musashi 1蛋白的表达情况,并分析结肠癌组织中Musashi 1蛋白和miRNA-132表达情况与患者临床特征的关系。结果结肠癌组织中Musashi 1蛋白的阳性表达率为68.75%,明显高于癌旁组织的28.75%,差异有统计学意义(P﹤0.01)。结肠癌组织中miRNA-132的相对表达量为(0.448±0.104),明显低于癌旁组织的(2.167±0.551),差异有统计学意义(P﹤0.01)。TNM分期为Ⅲ~Ⅳ期、有淋巴结转移、侵及浆膜的结肠癌患者结肠癌组织中Musashi 1蛋白的阳性表达率分别高于TNM分期为Ⅰ~Ⅱ期、无淋巴结转移、未侵及浆膜的结肠癌患者,差异均有统计学意义(P﹤0.05)。TNM分期为Ⅰ~Ⅱ期、高+中分化的结肠癌患者结肠癌组织中miRNA-132的相对表达量分别明显高于TNM分期为Ⅲ~Ⅳ期、低分化的结肠癌患者,差异均有统计学意义(P﹤0.01)。结论结肠癌组织中Musashi 1蛋白表达上调,miRNA-132表达下调,且其表达情况与结肠癌的恶性程度有关。
Objective To investigate the expression of Musashi 1 protein and microRNA-132(miRNA-132)in colon cancer tissues and the relationship with clinical characteristics.Method Eighty colon cancer specimens and the adjacent normal tissue specimens were collected.miRNA-132 and Musashi 1 protein expression were detected by quantitative realtime polymerase chain reaction(qRT-PCR)and immunohistochemical staining,the relationship between their expression and patients’clinical characteristics was analyzed.Result The positive expression rate of Musashi 1 protein in colon cancer tissues was 68.75%,which was significantly higher than that in adjacent normal tissues at 28.75%(P<0.01).The relative expression of miRNA-132 in colon cancer tissues was(0.448±0.104),which was significantly lower than that in adjacent normal tissues at(2.167±0.551)(P<0.01).The positive expression rate of Musashi 1 protein in colon cancer tissues with TNM stage III-IV,positive lymph node metastasis and serosal invasion was evidently higher compared to those with TNM stage Ⅰ~Ⅱ,negative lymph node metastasis,and no serosal invasion,the differences were of statistically significance(P<0.05).TNM stage Ⅰ~Ⅱ,high or moderate differentiation were associated with higher relative expression of miRNA-132 as compared with TNM stage Ⅲ~Ⅳ,and poor differentiation among colon cancer patients(P<0.01).Conclusion The up-regulated expression of Musashi 1 and down-regulated expression of miRNA-132 are present in colon cancer tissues,which are related to the degree of malignancy of tumors.
