摘要
目的探讨骨髓间充质干细胞(bone marrow mesenchymal stem cells,BMSCs)及其外泌体(BMSCs-exosomes,BMSCs-exo)对自发性高血压大鼠(spontaneous hypertensive rats,SHR)脑卒中的干预作用。方法体外分离法获取大鼠BMSCs诱导形成BMSCs-exo,然后尾静脉注射干预SHR,观察大鼠脑卒中发生率及患病时间。实验动物选取雄性12周龄SHR,随机分为BMSCs组、BMSCs-exo组和NaCl组,分别通过尾静脉注射BMSCs、BMSCs-exo和生理盐水干预SHR,Nor组为尾静脉注射生理盐水进行干预SD大鼠,测量各组大鼠体重、血压并观察大鼠生活习性,进行脑卒中临床评分;第6周,采用多普勒血流测量仪检测大鼠脑血流灌注量记录图像及数值;左心室穿刺法Medlab生理测量仪测定各组大鼠左心室压力,收集图像并记录。结果SHR经过6周干预处理,大鼠的尾动脉压和左心室压力BMSCs-exo组和BMSCs组较NaCl组大鼠明显降低,仍高于Nor组水平(P<0.05)。NaCl组大鼠脑血流灌注量显著低于Nor组(P<0.001),BMSCs-exo组和BMSCs组干预后大鼠脑血流灌注量增加(P<0.05),但2组间差异比较无统计学意义(P>0.05)。结论BMSCs和BMSCs-exo干预SHR,均可一定程度上降低大鼠外周血压及左心室压力,改善大鼠脑血流灌注量,减少SHR脑卒中发生率。
Objective To investigate the intervention effect of bone marrow mesenchymal stem cells(BMSCs)and BMSCs-exosomes(BMSCs-exo)on stroke in spontaneously hypertensive rats(SHR).Methods BMSCs was induced to form BMSCs-exo in vitro,and then injected into the SHR by tail vein.The incidence and duration of stroke were observed.The experimental animals selected male SHR of 12 weeks old were randomly divided into group BMSCs,BMSCs-exo group and NaCl group.BMSCs and BMSCs-exo and normal saline were injected into the tail vein to interfere with SHR.The Nor group was injected with saline in the tail vein to intervene the SD rats.The weight of the rats were measured,blood pressure was observed and the living habits of rats were observed,and then count the score of stroke.In the sixth weeks,the image and value of cerebral blood flow perfusion in rats were recorded by Doppler blood flow meter.Left ventricular puncture method throught Medlab physiological measuring instrument was used to measure the left ventricular pressure of rats in each group,and the images were collected and recorded.Results The SHR were treated for 6 weeks.The pressure of tail artery and left ventricular pressure in group BMSCs-exo and BMSCs group were significantly lower than that of group NaCl,which was still higher than that of group Nor(P<0.05).The cerebral blood flow perfusion in the NaCl group was significantly lower than that in the Nor group(P<0.001).The cerebral blood flow perfusion in the group BMSCs-exo and BMSCs group increased(P<0.05),and there was no significant difference between the two groups(P>0.05).Conclusion BMSCs and BMSCs-exo intervention in SHR can reduce peripheral blood pressure and left ventricular pressure and improve cerebral blood flow perfusion and then reduce the incidence of stroke in SHR.
