摘要
为了评价黄芪多糖作为疫苗佐剂的潜力,试验用黄芪多糖和灭活的H9N2亚型禽流感病毒免疫C57BL/6小鼠,将20只小鼠均分为4组,第1组为黄芪多糖组(免疫黄芪多糖),第2组为灭活H9N2亚型禽流感病毒组(免疫灭活H9N2亚型禽流感病毒),第3组为混合组(免疫黄芪多糖和灭活H9N2亚型禽流感病毒),第4组为生理盐水组(注射生理盐水),通过淋巴细胞增殖试验和流式细胞术对C57BL/6小鼠外周血淋巴细胞的增殖反应、B淋巴细胞和T淋巴细胞的变化及T淋巴细胞亚群比值变化进行研究。结果表明:混合组对小鼠外周血淋巴细胞的增殖有促进作用,可以提高小鼠外周血B淋巴细胞和CD^+_3CD^+_4T淋巴细胞亚群比例,并对小鼠外周血T淋巴细胞亚群有调节作用。说明灭活H9N2亚型禽流感病毒与黄芪多糖混合时具有协同作用。
In order to evaluate the immune adjuvant effect of astragalus polysaccharides on inactivated Avian influenza virus,C57BL/6 mice were immunized with astragalus polysaccharides and inactivated H9N2 subtype Avian influenza virus.Twenty C57BL/6 mice were divided into four groups,the first group was astragalus polysaccharides group(immunedby astragalus polysaccharides),the second group was inactivated H9N2 subtype Avian influenza virus group(immuned by inactivated H9H2 subtype Avian inflvenza virus),the third group was mixed group(immunedby astragalus polysaccharides and inactivated H9N2 subtype Avian influenza virus),the fourth group was saline group(injected with saline).Through lymphocyte proliferation test and flow cytometry,the proliferation response of peripheral blood lymphocytes,the change of B lymphocyte and T lymphocyte and the change of T lymphocyte subsets ratio in C57BL/6 mice were studied.The results showed that the mixed treatment could promote the proliferation of peripheral blood lymphocyte,increase the proportion of B-lymphocyte and CD+3 CD+4 T lymphocyte subpopulation,and regulate the T lymphocyte subpopulation in peripheral blood of mice.It indicated that the inactivation of H9N2 subtype Avi an influenza virus had a synergistic effect when it is mixed with astragalus polysaccharide.
