妊娠期糖尿病患者血清中TGF-β1表达水平及其基因rs1800470T/C位点单核苷酸多态性分析 被引量:1

摘要 目的了解妊娠期糖尿病(GDM)患者转化生长因子-β1(TGF-β1)水平情况,并探讨其基因rs1800470T/C位点单核苷酸多态性(SNP)与深圳地区GDM之间的遗传易感性。方法选择2017年6月至2019年5月于深圳市龙华区人民医院就诊并确诊为GDM患者154例(GDM组),年龄23~42岁,平均年龄29.03岁;孕周24~28周,平均孕周26.02周;体质量指数24~31 kg/m2。同期产检的非GDM孕妇120例(HC组),年龄24~40岁,平均年龄28.49岁;孕周25~28周,平均孕周26.53周;体质量指数23~32 kg/m2。采用酶联免疫吸附分析(ELISA)法检测TGF-β1水平,同时采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术检测TGF-β1基因rs1800470T/C位点SNP。结果 GDM组患者血清中TGF-β1水平为(23.97±3.48) ng/L,明显高于HC组[(14.06±1.52) ng/L],差异有统计学意义(t=3.105 7,P <0.05)。GDM组患者TGF-β1基因rs1800470T/C位点CC基因型和C等位基因检出率分别为31.17%和53.57%,明显高于HC组(12.50%和33.33%),差异有统计学意义(P <0.05);携带TGF-β1基因rs1800470T/C位点CC基因型GDM患者TGF-β1水平为(35.92±5.16) ng/L,明显高于TT和TC基因型[(17.57±1.89) ng/L和(19.07±2.46) ng/L],差异有统计学意义(t=4.260 3~4.528 9,P <0.05);而TT和TC基因型之间差异无统计学意义(t=0.980 5,P> 0.05)。结论GDM患者TGF-β1基因rs1800470T/C位点CC基因型及C等位基因检出率明显升高,且CC基因型GDM患者TGF-β1水平明显升高。因此,CC基因型可能是深圳地区GDM发病的易感危险遗传基因之一。 Objective To study the transforming growth factor-β1(TGF-β1)level in patients with gestational diabetes mellitus(GDM),and discuss susceptibility between its gene rs1800470T/C loci single nucleotide polymorphism(SNP)and GDM patients in Shenzhen.Methods From June 2017 to May 2019,154 GDM patients(GDM group)were enrolled,which aged 23-42 years old with mean age of 29.03 years old;gestational 24-28 weeks with mean gestational of 26.02 weeks;body mass index was24-31 kg/m2.In the same period,120 pregnant without GDM were set as HC group,which aged 24-40 years old with mean age of 28.49 years old;gestational 25-28 weeks with mean gestational of 26.53 weeks;body mass index was 23-32 kg/m2.The enzyme-linked immunosorbent assay(ELISA)method was used to detect TGF-β1level,and polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)technology in SNP of TGF-β1gene rs1800470T/C locus.Results The serum TGF-β1level in GDM group[(23.97±3.48)ng/L]was significantly higher than that in HC group[(14.06±1.52)ng/L],and difference was statistically significant(t=3.105 7,P<0.05).The detection rate of CC genotype and C allele of TGF-β1gene rs1800470T/C locus in GDM group(31.17%and 53.57%)was significantly higher than that in HC group(12.50%and33.33%),and difference was statistically significant(P<0.05);The GDM with TGF-β1gene rs1800470T/C locus CC genotype[(35.92±5.16)ng/L]was significantly higher than that of TT and TC genotypes[(17.57±1.89)ng/L and(19.07±2.46)ng/L],and difference was statistically significant(t=4.260 3-4.528 9,P<0.05);There was no significant difference between TT and TC genotypes(t=0.980 5,P>0.05).Conclusion It is demonstrated that detection rate of CC genotype and C allele of TGF-β1gene rs1800470T/C locus in GDM patients is significantly increased,and TGF-β1level in GDM patients CC genotype is significantly increased.Therefore,CC genotype may be one of the susceptible genes for GDM in Shenzhen.
