摘要
目的探讨复方生地合剂治疗系统性红斑狼疮(SLE)的可能作用机制。方法将30只MRL/lpr小鼠(狼疮模型鼠)随机分组为模型组、复方生地合剂组、强的松组,每组10只,另以10只C57BL/6小鼠为空白组。复方生地合剂组给予浓度为2.8g/ml的复方生地合剂人临床等效剂量18.495ml/(kg·d)灌胃,强的松组给予浓度为1.23g/ml强的松片混悬液10ml/(kg·d)灌胃,模型组和空白组给予生理盐水10ml/(kg·d)灌胃,各组均每日1次。连续灌胃8周后检测血清白细胞介素17A(IL-17A)、转化生长因子β1(TGF-β1)水平,检测脾脏淋巴细胞中辅助性T细胞17(Th17)、调节性T细胞(Treg)水平和脾脏组织维甲酸相关孤核受体(RORγt)和叉头状转录因子p3(Foxp3) mRNA表达。试验期间观察各组小鼠死亡情况。结果复方生地合剂组死亡2只,强的松组死亡1只,模型组死亡3只,空白组无死亡。与空白组比较,模型组小鼠血清IL-17A水平、脾脏组织RORγt mRNA表达升高,脾脏淋巴细胞Th17/CD4^+、Th17/Treg值亦升高,血清TGF-β1水平、脾脏组织Foxp3 mRNA表达、Treg/CD4^+值降低(P<0.05或P<0.01)。与模型组比较,复方生地合剂组和强的松组小鼠血清IL-17A水平、RORγt mRNA表达、Th17/CD4^+、Th17/Treg值降低,血清TGF-β1水平、Treg/CD4^+值升高(P<0.05或P<0.01)。复方生地合剂组和强的松组各指标比较差异无统计学意义(P>0.05)。结论复方生地合剂可以改善脾脏组织Th17/Treg平衡,可能是其治疗SLE的作用机制之一。
Objective To explore possible mechanism of Compound Shengdi Mixture(复方生地合剂) in treatment of systemic lupus erythematosus(SLE). Methods Totally 30 MRL/lpr mice(lupus model mice) were randomly divided into model group, Compound Shengdi Mixture group and prednisone group, with 10 mice in each group, and 10 C57 BL/6 mice were considered as blank group. The Compound Shengdi Mixture group was given 2.8 g/ml Compound Shengdi Mixture with a human clinical equivalent dose of 18.495 ml/(kg·d) by gavage. The prednisone group was given 10 ml/(kg·d) 1.23 g/ml prednisone suspension by gavage. The model group and the blank group were given 10 ml/(kg·d) normal saline by gavage, once a day for each group. After 8 weeks of conti-nuous intragastric administration, interleukin-17 A(IL-17 A) and transforming growth factor-β1(TGF-β1) levels in serum were determined. Helper T cell 17(Th17) and regulatory T cell(Treg) levels in splenic lymphocytes, and mRNA expressions of retinoid acid orphan receptor γt(RORγt) and fox head box protein p3(Foxp3) in splenic tissue were determined. Death of the mice in each group was observed during the experiment. Results There were 2 mice dead in the Compound Shengdi Mixture group, 1 in the prednisone group, 3 in the model group and 0 in the blank group. Compared to the blank group, IL-17 A level in mice serum and the mRNA expression of RORγt in splenic tissue in the model group were increased;values of Th17/CD4^+ and Th17/Treg in splenic lymphocytes were also increased;TGF-β1 level in serum, the mRNA expression of Foxp3 in splenic tissue and the value of Treg/CD4^+ were decreased(P<0.05 or P<0.01). Compared with the model group, the IL-17 A levels in mice serum, the mRNA expressions of RORγt, as well as values of Th17/CD4^+ and Th17/Treg in the Compound Shengdi Mixture group and the prednisone group were decreased, and the TGF-β1 level in serum and the value of Treg/CD4^+ were increased(P<0.05 or P<0.01).
