萝卜硫素通过Nrf2/ARE通路缓解矽肺模型大鼠肺组织纤维化及氧化应激的实验研究 被引量:14

摘要 目的研究萝卜硫素对矽肺模型大鼠肺组织纤维化、氧化应激及抗氧化通路核因子E2相关因2(Nrf2)/抗氧化反应元件(ARE)通路的影响。方法选择SD大鼠并随机分为对照组、模型组和萝卜硫素组,对照组支气管灌注生理盐水,后两组给予1 ml含有50 mg/ml二氧化硅的生理盐水支气管灌注以建立矽肺模型。造模当天开始,萝卜硫素组给予5 mg/kg萝卜硫素腹腔注射,对照组和模型组给予等剂量生理盐水腹腔注射,1次/d,连续8周。取肺组织进行HE染色并检测胶原蛋白、氧化应激介质的含量及Nrf2/ARE通路分子的表达量。结果与对照组比较,模型组大鼠HE染色可见明显的病理变化且肺组织中胶原蛋白-I(Col-I)、胶原蛋白-III(Col-III)、丙二醛(MDA)的含量及Nrf2、ARE的表达量明显增多,超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH)的含量及Kelch样环氧氯丙烷相关蛋白-1(Keap-1)的表达量明显减少,差异均有显著性(P<0.05);与模型组比较,萝卜硫素组大鼠HE染色可见病理变化减轻且肺组织中Col-I、Col-III、MDA的含量及Keap-1的表达量减少,SOD、GSH的含量及Nrf2、ARE的表达量明显增多,差异均有显著性(P<0.05)。结论萝卜硫素能够缓解矽肺模型大鼠肺组织纤维化及氧化应激,且该作用与Nrf2/ARE通路的激活有关。 Objective To study the effects of sulforaphane on pulmonary fibrosis,oxidative stress and nuclear factor erythroid 2-related factor 2(Nrf2)/antioxidant response element(ARE)pathway in silicosis rats.Methods SD rats were randomly divided into control group,model group and sulforaphane group.The control group was given bronchial saline.The latter two groups were given bronchial perfusion of physiological saline containing 50 mg/m L silica to establish silicosis model.The sulforaphane group was given 5 mg/kg sulforaphane intraperitoneal injection,control group and model group were given intraperitoneal injection of saline,once a day for 8 weeks.Lung tissues were stained with HE and the contents of collagen,oxidative stress media and the expression of Nrf2/ARE pathway molecules were detected.Results Compared with the control group,HE staining showed significant pathological changes,and the contents of Collagen-I(Col-I),Collagen-III(Col-III),malondialdehyde(MDA)and the expression of Nrf2,ARE in lung tissue significantly increased,while the contents of superoxide dismutase(SOD),glutathione peroxidase(GSH)and the expression of Kelch-like ECH-associated protein 1(Keap-1)significantly decreased in the model group.Compared with the model group,HE staining showed pathological changes alleviated and the contents of Col-I,Col-III,MDA and the expression of Keap-1,while the expression of SOD,GSH and the expression of Nrf2,ARE significantly increased in lung tissue in the sulforaphane group.Conclusion Sulforaphane can alleviate pulmonary fibrosis and oxidative stress in silicosis rats,and this effect is related to the activation of Nrf2/ARE pathway.
