摘要
为了筛选出适合甜菜分子标记的InDel引物以及利用InDel引物构建能够扩增多重InDelPCR反应的引物,以提高甜菜InDel-PCR扩增的效率,利用8个甜菜品种的DNA对300对甜菜InDel引物进行筛选,结果筛选出多态性较好、条带清晰、易于识别的甜菜InDel引物44对,这44对引物共扩增出总条带116条,其中多态性条带109条,多态性比率为94%;其中有37对引物扩增的总条带为2或3条,可以作为将来多重InDel引物的首选;又从中筛选出7对多重InDel引物:ND228、ND31、ND267、ND229、ND66、ND231、ND22。
In order to select the primers suitable for sugar beet molecular marker InDel and to construct the primers which can amplify the multiple InDel-PCR reaction for improving the efficiency of the InDel-PCR amplification.A total of 300 InDel primers were screened using the DNA of eight beet varieties,and 44 primer pairs of sugar beet InDel with good polymorphism,clear bands and easy to identify were selected.These 44 pairs of primers amplified a total band of 116,among which,109 were polymorphic bands with a polymorphism ratio of 94%,and 37 pairs of primers amplified a total band of 2 or 3,which could be considered as the first choice for multiple InDel primers in the future;and 7 pairs of multiple InDel primers ND228,ND31,ND267,ND229,ND66,ND231 and ND22 were selected.
