摘要
目的探讨分析干扰素-γ(IFN-γ)基因多态性与脊柱结核易感性及与血清白细胞介素-4(IL-4)、IFN-γ的关系。方法选择2016年4月-2018年9月杭州市红十字会医院收治的脊柱结核患者78例作为脊柱结核组,同期健康志愿者100名作为对照组,采用聚合酶链反应-限制性片段长度多态性分析技术比较两组研究对象IFN-γrs2430561位点基因多态性,并对比两组受试者血清IL-4、IFN-γ水平。结果脊柱结核组患者IFN-γrs2430561位点A、T两个等位基因频率分别为88.46%(138/156)、11.54%(18/156),而对照组为80.00%(160/200)、20.00%(40/200),两组差异有统计学意义(P<0.05)。脊柱结核组患者血清IFN-γ为(14.37±3.71)ng/L高于对照组(P<0.001),而两组IL-4差异无统计学意义(P=0.520)。脊柱结核IFN-γrs2430561位点AA基因型患者血清IFN-γ为(13.79±2.42)ng/L较AT+TT基因型低(P=0.025),而各基因型患者血清IL-4差异无统计学意义。结论IFN-γrs2430561位点基因多态性与脊柱结核有关,A等位基因作为脊柱结核的易感基因,该位点基因多态性可影响宿主IFN-γ表达,从而发挥结核免疫保护调控作用。
OBJECTIVE To investigate the association of IFN-gamma gene polymorphism with susceptibility to spinal tuberculosis and serum of IL-4 and IFN-γ.METHODS Seventy-eight patients with spinal tuberculosis admitted to Hangzhou Red Cross Hospital hospital from Apr.2016 to Sep.2018 were selected as the spinal tuberculosis group,and 100 healthy volunteers as the control group.Polymerase chain reaction-restriction fragment length polymorphism(PCRRFLP)analysis was used to compare the gene polymorphism of IFN-γrs2430561 locus between the two groups,and the serum IL-4 and IFN-γlevels in the two groups of subject were compared.RESULTS The frequencies of IFN-gamma rs2430561 A and T alleles were 88.46%(138/156)and 11.54%(18/156)in the patients with spinal tuberculosis,respectively,compared with 80.00%(160/200)and 20.00%(40/200)in the control group,with significant difference between the two groups(P<0.05).The serum IFN-gamma level in patients with spinal tuberculosis was(14.37±3.71)ng/L,significantly higher than that in control group(P<0.001),while there was no significant difference of IL-4 between the two groups(P=0.520).The serum IFN-gamma level of AA genotype at IFN-gamma rs2430561 locus in spinal tuberculosis patients was(13.79±2.42)ng/L,significantly lower than that of AT+TT genotype(P=0.025),while there was no significant difference of serum IL-4 among different genotypes.CONCLUSION The polymorphism of IFN-γrs2430561 locus is closely related to the susceptibility of spinal tuberculosis.As a susceptibility gene of spinal tuberculosis,the A allele polymorphism of IFN-γlocus could affect the expression of IFN-γin host and play a role in regulating and protecting tuberculosis immunity.
