猪繁殖与呼吸综合征病毒NADC30-like毒株的分离鉴定 被引量:3

Isolation and Identification of a Porcine Reproductive and Respiratory Syndrome Virus NADC30-like Strain

摘要 对河南漯河地区某养殖场检出的猪繁殖与呼吸综合征病毒(PRRSV)阳性病料进行处理,接种到原代猪肺泡巨噬细胞中,经细胞盲传成功分离出1株PRRSV毒株,命名为HeNLH2017株,并测定了该分离株在原代猪肺泡巨噬细胞上的生长曲线。为进一步分析该分离株的遗传进化特点,扩增测定了ORF5基因和部分Nsp2基因的核酸序列。核酸同源性分析显示,该分离毒株ORF5基因和Nsp2序列与NADC30毒株同源性最高,分别为93.4%和93.8%。遗传进化树分析显示,该分离株与我国2013年以来流行的NADC30-like(Lineage1)毒株处于同一进化分支。氨基酸序列比对分析显示,ORF5基因和Nsp2序列与NADC30-like毒株具有相似的遗传变异特点,且Nsp2序列存在特征性的131个不连续的氨基酸缺失。成功分离到1株NADC30-like毒株,有助于进一步认识NADC30-like毒株在河南地区的流行情况和临床致病特点。 A PRRSV strain was successfully isolated from clinical PRRSV-positive samples collected from Luohe,Henan province and named as HeNLH2017.The growth curve was determined on porcine alveolar macrophages(PAMs).To further analyze the genetic evolution,we amplified the nucleic acid sequences of the ORF5 gene and highly variable region of the Nsp2 gene.The homology analysis showed that the ORF5 gene and partial Nsp2 sequence had the highest homology with the NADC30 strain,which were 93.4%and 93.8%,respectively.Genetic phylogenetic tree analysis showed that the isolate was clustered into the same evolutionary branch with the NADC30-like(lineage1)strain that has been epidemic since 2013 in China.Amino acid sequence alignment showed that the ORF5 gene and Nsp2 sequence have similar genetic variation characteristics to the NADC30-like strain,and the Nsp2 sequence has characteristic 131 discontinuous amino acids deletion.In brief,we successfully isolated a NADC30-like strain,which is helpful to further understand the prevalence and clinical pathogenic characteristics of NADC30-like strain in Henan.
