霉酚酸酯对过敏性紫癜小鼠T-bet、GATA3表达及Th1/Th2平衡的影响 被引量:6

Effects of Mycophenolate Mofetil on T-bet and GATA3 Expressions and Th1/Th2 Balance in Mice with Allergic Purpura

摘要 目的探讨霉酚酸酯对过敏性紫癜小鼠T-bet、GATA3表达及Th1/Th2平衡的影响。方法建立过敏性紫癜小鼠模型60只,随机分为模型组、霉酚酸酯低(100mg/kg)、中(200 mg/kg)、高(300 mg/kg)剂量组、西咪替丁组(1 g/kg),另取12只BALB/c小鼠设为对照组,药物处理组于建模成功后每天灌胃治疗,持续3周。末次给药24 h后处死小鼠,采用印度墨水小鼠碳粒廓清法检测各组小鼠单核巨噬细胞系统(MPS)功能,计算吞噬系数(K值)与吞噬指数(α值),采用微量聚乙二醇沉淀法测定各组小鼠血清循环免疫复合物(CIC)含量,采用酶联免疫吸附试验(ELISA)检测各组小鼠血清中干扰素γ(IFN-γ)、白介素-2(IL-2)、白介素-4(IL-4)、白介素-10(IL-10)的水平,以流式细胞术检测各组小鼠外周血中T淋巴细胞Th1/Th2比值,以免疫印迹法(Western blot)检测各组小鼠外周血单核细胞中蛋白T-bet、GATA3的表达情况。结果与对照组比较,模型组小鼠K值、α值、IFN-γ、IL-2水平、Th1/Th2比值、T-bet蛋白表达均明显降低(P<0.05),IL-4及IL-10水平、CIC含量、GATA3蛋白表达均明显升高(P<0.05);与模型组比较,药物处理组小鼠K值、α值、IFN-γ、IL-2水平、Th1/Th2比值、T-bet蛋白表达均升高(P<0.05),IL-4及IL-10水平、CIC含量、GATA3蛋白表达均降低且霉酚酸酯各组呈剂量依赖性(P<0.05);霉酚酸酯高剂量组与西咪替丁组相比差异无统计学意义(P>0.05)。结论霉酚酸酯可能通过调控T-bet/GATA3表达,恢复过敏性紫癜小鼠Th1/Th2平衡,减轻过敏性紫癜小鼠症状。 Objective expression and Th1/Th2 balance in mice with allergic purpura.Methods model mice with allergic purpura were established and randomly divided into model group,low-(100 mg/kg),medium-(200 mg/kg),high-(300 mg/kg)dose mycophenolate mofetil groups and cimetidine group(1 g/kg),and another 12 BALB/c mice were selected as control group.The drug treatment group was given intragastric administration every day for three weeks after successful modeling.The mice were sacrificed at 24 h after the last administration.The function of mononuclear macrophage system(MPS)was detected by carbon particle clearance method in Indian ink,and the coefficient of phagocytosis(K value)and index of phagocytosis(αvalue)were calculated.In addition,the content of circulating immune complex(CIC)in serum was determined by micropolyethylene glycol precipitation method,and the levels of serum interferon-γ(IFN-γ),interleukin-2(IL-2),interleukin-4(IL-4)and interleukin-10(IL-10)were measured by enzyme-linked immunosorbent assay(ELISA).T lymphocyte Th1/Th2 ratio in peripheral blood was detected by flow cytometry,and the expressions of T-bet and GATA3 in peripheral blood mononuclear cells of mice was detected by western blot in each group.Results control group,K value,αvalue,IFN-γ,IL-2 level,Th1/Th2 value and T-bet protein expression in model group were significantly decreased(P<0.05),and IL-4 and IL-10 levels,CIC content and GATA3 protein expression were significantly increased(P<0.05).Compared with model group,K value,αvalue,IFN-γ,IL-2 level,Th1/Th2 value and T-bet protein expression in drug-treated mice were increased(P<0.05),while IL-4 and IL-10 levels,CIC content and GATA3 protein expression were decreased in a dose-dependent manner between mycophenolate mofetil groups(P<0.05).There was no significant difference between high-dose mycophenolate mofetil group and cimetidine group(P>0.05).Conclusion Mycophenolate mofetil may restore Th1/Th2 balance and alleviate the symptoms of mice with allergic purpura by regulating the expression of
